Backgroud&Objective:Endometriosis (EMs) is one of common refractory benign diseases of gynecology.Endometriosis may affect6-10%of women of reproductive age. At present, Laparoscopic surgery is the gold standard for the diagnosis of endometriosis.Telomeres are located at the ends of eukaryotic chromosomes and essential for chromosomes stability. The length of telomere plays important role to the control of cell mitosis. Telomerase is of great significance for maintaining telomere length, By activation of this enzyme,cells are able to overcome replicative senescence and to divide indefinitely.Human telomerase reverse transcriptase (hTERT) is the catalytic subunit of telomerase, which is crucial for the activity of telomerase. A large numbers of studies prove that the expression of hTERT in tumor tissues is high, however, the studies on expression of hTERT in endometriosis is rare.The objective of this thesis is to investigate the expression of hTERT in the eutopic and ectopic endometrium of women who is diagnosed with endometriosis and appraise the role of hTERT in the pathogenesis of endometriosis.Method:In this study, the expression of hTERT was evaluated in the endometrial specimens of54women diagnosed with endometriosis and of15healthy women by immunohistochemistry.To assess the presence of hTERT in both eutopic and ectopic endometrium of women with endometriosis, then subsequently compared with normal endometrium.At last,we analysed the relishenship between the expression of hTERT in EMs and CA125,r-AFS stage,age.Results:In the eutopic and ectopic endometrium of women with endometriosis,immunoreactivities of hTERT increased compared with those of normal endometrium (p<0.05). In the ectopic endometrium of EMs,the immunoreactivities of hTERT increased compared with the eutopic endometriosis of EMs(p<0.05).We also observed a cyclic variation in the eutopic endometrium with and without EMs throughout their menstrual cycle,the presence of hTERT in endometrial cells has no different with the menstrual cycle (p>0.05). The expression of hTERT in the eutopic endometrium of EMs is higher than the expression of hTERT in the normal endometrium in the proliferative phase (p<0.05).However, there is no obvious diference between the expression of hTERT in the eutopic endometrium and the expression of hTERT in the normal endometrium in the secretory phase (p>0.05).The presence of hTERT in EMs has positive correlation with CA125(p<0.05,r>0).There is no correlation between the presence of hTERT and r-AFS stage (p>0.05) and age (p>0.05)Conclusion:These findings indicate that hTERT may be involved in the process of endometriosis.The abnormal proliferation of endometrium in endometriosis may play a role in the happing of the disease.The combination of hTERT immunohistochemistry and serum CA125may evaluate the malignant of endometriosis.
