| Objective: Esophageal cancer is one of the world’s top ten tumor, with a high incidence inmales, especially. In our country, the main pathological type is esophageal squamous cellcarcinoma (ESCC). Looking for the early and prognosis markers of ESCC could help theearly diagnosis and accurately classify prognostic risks for patients. Methods: This studycollected the specimens of442cases of ESCC tissues and150cases of precancerouslesions in esophagus. We examined the expression of20proteins by tissue microarrayimmunohistochemical technique(TMA-IHC), and also analyzed the correlation betweenthe overexpression of these proteins and clinical pathological parameters. Results: Of20proteins tested, AHNAK2, TNS4, N-cadherin, TP53and MRE11showed high frequencyexpression in carcinoma tissues, whereas none or low detectable in esophageal adjacentnormal tissues. As to AHNAK2, the rate of negative, weak, moderate and high positiveexpression were24.8%,28.7%,17.0%and29.5%in tumors, respectively. Kaplan-Meiersurvival curves showed that ESCC patients with negative expression of AHNAK2hadshorter overall survival time (P﹤0.05). Positive expression of AHNAK2was closelyrelated to tumor differentiation (P﹤0.01). Negative, weak, moderate and high expressionof TNS4were observed in37.0%,25.3%,16.6%and21.1%of tumors, respectively. Theresults showed that: Positive expression of TNS4was significantly correlated with tumordifferentiation (P﹤0.01), T stage(P﹤0.01) and TNM stage(P﹤0.05). Negative,weak, moderate and high positive expression rate of N-cadherin were observed in7.5%,10.0%,42.5%and40.0%of tumors, respectively. Positive expression of N-cadherin isassociated with tumor differentiation(P﹤0.05). The positive expression rate of the TP53and MRE11protein is very low and with a high specificity in paracancerous tissues.Further experiments in precancerous tissues, the results is that TP53protein in normal group, mild dysplasia, moderate dysplasia, severe atypical hyperplasia tissues in thepositive rate is4.9%,22.2%,40.0%and29.2%, respectively. We also found that theexpression of TP53protein in normal tissue and mild to moderate dysplasia organizationhas significant difference (P﹤0.01), the expression of TP53protein in normal and severeatypical hyperplasia tissues was also has a obvious statistical significance (P﹤0.05).MRE11protein in the normal group, mild dysplasia, moderate dysplasia, severe dysplasiain the positive expression rate of3.1%,26.7%,3.1%and26.7%., respectively. Theexpression of MRE11in normal and severe dysplasia tissues have a significant difference(P﹤0.01), the expression of MRE11in mild dysplasia and severe dysplasia tissues wasalso statistically significant (P﹤0.05). Conclusion: TNS4, N-cadherin and AHNAK2expression might contribute to the prognostic evaluation of patients with this disease.TP53, MRE11might be associated with the early diagnosis. |
