| Objective To investigate the stage of rats’ gastric ulcer, the restored mucosa thickness,detect PGE2, EGF and EGFR in the gastric mucosa, detect SS, CGPR,TFF3and NO inthe blood, and approach the affection and mechanism of gastric ulcer healing inexperiment acetic acid induced in rats under+Gz condition.Methods32male Sprague-Dawley rats were randomly and equally divided into acontrol group, a+5Gz exposure group, a+10Gz exposure group, and+10Gz gavagedwith KangFuXinYe solution group. Gastric ulcer was induced in rats with acetic acid.After3d, all the rats in four groups were exposed to+Gz each other day for1wk (fourtimes in all, five minutes each time). The rats in+10Gz gavaged with KangFuXinYesolution group were gavaged with KangFuXinYe solution2milliliter every day for1week. Then executed all the rats, observed stage of gastric ulcer and the ultrastructure ofregenerative mucosa by electronic microscopy, tade tissue and bloods samples to detectthe contents of prostaglandin E2(PGE2), Epidermal growth factor (EGF) andEpidermal growth factor receptor(EGFR) in the gastric mucosa, ghrelin,somatostatin(SS), calcitonin gene-related peptide (CGRP),and trefoil peptide(TFF3) inblood by radioimmunoassay, and detected the contents of nitric oxide (NO) in blood bynitrate reductase assay.Results (1) The observation of macropathology: The higher+Gz, the lower stage of thegastric ulcer, and there were serious hyperemia and hydropic degeneration.The qualityof ulcer healing in+10Gz gavaged with KangFuXinYe solution group was better thanthe+10Gz group.(2) The stage of gastric ulcer: gastric ulcers of control group inrepair stage were major, accounted5/8, gastric ulcers of+5Gz group in scarring stageaccounted4/8, gastric ulcer of+10Gz group in acute stage accounted5/8, and gastric ulcer of+10Gz gavaged with KangFuXinYe solution group in repaire stage accounted5/8, more than the gastric ulcer of+10Gz group.(3) The higher+Gz, the thinnerrestored mucosa thickness and the more the number of cystically dilated glands underelectronic microscopy. The mucosa glands in+10Gz gavaged with KangFuXinYesolution group were more regular than+10Gz group.(4) The contents of PGE2in thegastric mucosa were significantly lower in the+10Gz exposure group than in the+5Gzexposure group, control group (3.438±0.908pg/mg vs5.15±0.65pg/mg vs6.99±0.74pg/mg)(P<0.01). The PGE2contents of+10Gz group were lower than+10Gz gavagedwith KangFuXinYe solution group(3.44±0.91pg/mg vs6.13±0.55pg/mg)(P<0.05).(5)The contents of CGRP in blood were significantly lower in the+10Gz exposure groupthan in the+5Gz exposure group, control group (62.25±15.94pg/ml vs78.33±11.29pg/ml vs82.58±11.79pg/ml)(P<0.05). The contents of NO in blood were significantlylower in the+10Gz exposure group than in the+5Gz exposure group, control group(44.77±6.86μmol/l vs47.78±2.85μmol/l vs53.81±9.27μmol/l)(P <0.05).(5) TheEGF contents of gastric mucosa: The control group were0.14±0.01pg/mg,+5Gz groupwere0.10±0.03pg/mg,+10Gz group were0.12±0.04pg/mg.+5Gz group were lowerthan control group (P<0.05), there were no different among+10Gz group, controlgroup and+5Gz group(P>0.05). The EGFR contents of gastric mucosa: The controlgroup were6.91±0.98pmol/g,+5Gz group were5.55±1.23pmol/g,+10Gz group were6.19±1.47pmol/g.+5Gz group were lower than control group (P<0.05), and therewere no different between+10Gz group and+5Gz group(P>0.05).(7) The SS inbloods of+5Gz group was higher than control group and+10Gz group(51.52±10.88pg/ml vs38.37±14.16pg/ml vs32.65±11.68pg/ml, P<0.05), and+10Gz group werenot different from control group(P>0.05).The ghrelin in bloods of+10Gz group waslower than+5Gz group(94.48±23.96ng/ml vs142.56±38.63ng/ml, P<0.05), and therewere no significant difference between control group and+5Gz group(112.00±42.28ng/ml vs142.56±38.63ng/ml, P>0.05). The trefoil peptide in bloods of the three group were no sidnificant difference(P>0.05).Conclusions (1) The gastric ulcer of experiment rats delayed, and the paturity ofregenerative mucosa and the stage of gastric ulcer decreased under+Gz condition.(2)The+Gz increased, the contents of PGE2in gastric mucosa, the contents of CGFR andNO in bloods decreased, and the restoration of regenerative mucosa delayed.KangFuXinYe could promote the healing of gastric ulcer.(3) Under+5Gz explosed,EGF and EGFR contents in gastric mucosa decreased, but had not significanlydifference under+10Gz explosed.(4) Under+5Gz, the contents of SS increased, thecontents of ghrelin had no difference with control group; under+10Gz, both thecontents of SS and ghrelin decreased; under positive acceleration trefoil peptide didn’tparticipate the healing of gastric ulcer. |
