| Background and aim:Bladder Urothelial Carcinoma is a common malignancy of the urinary system, Invasion and metastasis is the most important factor of poor prognosis of bladder cancer, explore the mechanism of tumor invasion and metastasis, find the relevant factors for bladder cancer invasion and metastasis, and has important implications for the treatment and prognosis of bladder cancer.the role of epithelial-mesenchymal transformation in the tumor has been growing awareness, in this study, we investigate the expression of transcription factor Slug and epithelial cells cadherin E-cadherin(the key factors of epithelial-mesenchymal transformation) in bladder urothelial carcinoma tissue and normal tissue, and the difference between pathological grades and stages, then, verify the corelation between expression of Slug and E-cadherin.Methods:40cases of surgically resected fresh bladder urothellal carcinoma tissue and normal tissue were collected from2010-2012in the department of pathology, the NO1hospital of ShanXi Medical University. the patients had not been adjunctive therapy before operation Pathology and clinical data were all.The routine H&E-stained sections were reviewed to verify me morphologic diagnosis. RT-PCR analysis was used to detect the gene expression of Slug and E-cadherin. Westem-blot analysis was used to detect the protein expression of Slug and E-cadherin. Statistical analysis was performed using the SPSS16.0software.Results:The Slug mRNA and protein expression in the bladder urothellal carcinoma tissue was higher than that in the normal tissue(p<0.05). The E-cadherin mRNA and protein expression in the bladder urothellal carcinoma tissue was lower than that in the normal tissue(p<0.05). The expression of Slug and E-cadherin was significantly related with the differential clinical stage, pathological degree, and lymph node metastasis(p<0.05). The expressions of Slug and E-cadherin have no association with age, sex(p>0.05).There was negative correlation between expression of Slug and E-cadherin in bladder urothelial carcinoma(p<0.05).Conclusion:The high expression of Slug and low expression of E-cadherin in bladder urothelial carcinoma are involved in the processes of invasion and metastasis of bladder urothelial carcinoma, Epithelial-mesenchymal transformation to play an important role in the development of bladder urothelial carcinoma. |
