Purpose:To investigate the inhibitory effect and chemosensitization of gene recombinant plasmid pDC316-hIL-24used alone or in combination with cisplatin(DDP) on human cervical cancer nude mice tumor growth and metastasis.Methods:The cervical cancer cells (Siha cells) were inoculated subcutaneously into the female c/mice to establish xenograft models,then which were randomly divided into six groups, each group had6mice. As follows:the control group of PBS, empty plasmids pDC316group, recombinant plasmids pDC316-hIL-24, half dose DDP group, full dose DDP and recombinant plasmids pDC316-hIL-24+half dose DDP group. After treatment, observed and compared the nude mice general condition, the change of weight and the tumor growth. At the end of the trial, we killed the mice and took their vena ophthalmica blood, the tumors and splenic organs. Tumor growth inhibitory rate and spleen indexes were calculated. The expression of mRNA of IL-24in transplanted tumors was identified by PCR. The expression of TNF-α、IFN-γ were evaluated with ELISA technique. The variation trend of PCNA were detected by Western blotting technique. b-FGF、MVD were detected with immunohistochemistry technology.Results:1. pDC316-hIL-24was successfully transfected into and expressed in nude mice xenograft models.2. Compared with the other three groups, the average inhibitive rates of IL-24+half dose DDP group、IL-24group and full dose DDP (51.5%、33.9%、36.3%)were significantly higher than the remaining groups (P<0.001). Among6groups, IL-24+half dose DDP yielded higher inhibitive rates (P<0.001) and lower side effect, but the PBS groups and empty plasmids pDC316group had no significant different (P>0.05).3.The TNF-α、IFN-γ and spleen indexes of IL-24group and combined group had a significant higher then other groups. Oppositely, the group of full dose DDP had a significant reduction.4. The results of Western blotting and immunohistochemistry technology showed that the expressions of PCNA,b-FGF and MVD were significantly lower between IL-24and combined group(P<0.01).Conclusion:1. The combined intervention of pDC316-hIL-24and DDP in nude mice xenograft models with human cervical cancer Siha cells has significant inhibiting growth function.2. IL-24can enhance the chemosensitization of DDP, at the same time,reduce the side effect of single adjuvant chemotherapy.3. The antitumor mechanisms and the chemosensitization is likely IL-24can enhance body immune functions, inhibit tumor cells multiplication and micro vessel formation.
