1Objective: By investigating the effects of Pingchuanning on Bcl-2and Bax Proteinexpressions in cold asthma model rats, and the intervention of ERK signal pathway, toexplore the molecular mechanisms of prevention and treatment ofasthma withPingchuanning.To provide a theoretical basis for clinical treatment.2Method:105SD male rats were randomized into Normal group、model group、theGuilong kechuanning group、Dexamethasone group、Pingchuanning high-dose group、Pingchuanning dose group and Pingchuanning low-dose group with15in each.To copythe ovalbumin sensitized and cold stimulation in rat asthma model, Modeled after21days, normal, model control groups were given the same amount of distilled water everyday to fight the suit, the dexamethasone group, Guilong kechuanning group and high、medium and dose groups of Pingchuanning were given each drug according to humanand rat body surface area than the conversion operator. And observed the changes inlung function of asthma rats after the atomization of4weeks, then removed the lungtissue. Detected by immunohistochemical assay experiments the Bcl-2and Bax Proteinexpressions of the lung tissues; Using the RT-PCR assay experimental lung tissueMEK1mRNA、MEK2mRNA、RasmRNA expression abundance; Detected by ELISAexperiments lung tissue between the cGMP and cAMP in the ratio; observed thepathological changes of lung tissue by HE staining.3Result:3.1The comparison of Bcl-2and Bax in lung tissue measuring results: There isdifference between the model group and the normal group(p<0.01); compared with themodel group, the Bcl-2expression in the lung tissue of each treatment groups werereduced(p<0.05), and the Bax expression were increased(p<0.05); compared withDexamethasone group and Guilong kechuanning group, the Bcl-2expression in the lungtissue of Pingchuanning high-dose group was reduced(p<0.01), and the Bax in lungtissue of Pingchuanning high-dose group was increased(p<0.01). 3.2The comparison of MEK1mRNA、MEK2mRNA、RasmRNA in lung tissuemeasuring results: There is difference between the model group and the normalgroup(p<0.01); compared with the model group, the MEK1mRNA、MEK2mRNA、RasmRNA expression in the lung tissue of each treatment groups were reduced(p<0.05);compared with Dexamethasone group and Guilong kechuanning group, theMEK1mRNA、 MEK2mRNA、 RasmRNA expressoin in the lung tissue ofPingchuanning high-dose group were reduced(p<0.01).3.3The comparison of cAMP、cGMP in lung tissue measuring results: Pingchuanninghigh-dose group and Pingchuanning dose group for cAMP、cGMP content and rationhave Statistically significant(p<0.05,p<0.01).4Conclusion: Pingchuanning could degrede the airway hyperreactivity and improvethe lung function,then treat the asthma by adjusting the expression of Bcl-2protein andBax protein in lung of rats.
