Study On The Association Bettween TCM Syndromes Of Essential Hypertension And Polymorphism Of Angiotensinogen

Objective：1. By studying the distribution of the Polymorphism of seven genetic SNPs ofPatients with different TCM syndrome types of essential hypertension, finding theeffective genetic SNPs which affect the syndromes of essential hypertension, to revealthe essence of the syndromes of essential hypertension from the Point of view withmolecular genetics.2. Through the relevance research of different syndromes of essential hypertensionbetween the distribution of Polymorphism of angiotensinogen and the intermediatePhenotype, to discuss Part of the mechanism of the distribution characteristics betweenthe syndrome classification and the intermediate Phenotype.Methods：Choose four hundred Patients with essential hypertension, divide them with fourdifferent TCM syndromes, which are: Liver-fire hyperactivity type、Yin deficiency withyang hyperactivity type、Phlegm-damp retention type、dual Deficiency of yin and yangtype, one hundred in each type, then select one hundred normal Persons for the controlgroup, collect their samples of blood, test their Polymorphism of angiotensinogen withthe technique called Polymerase chain reaction, and the Plasma levels ofangiotensinogen with the method of Enzyme Linked Immuno Sorbent Assay, Input allthe information of Patients and the testing data to the excel spreadsheet, then builddatabases, with SPSS to do the data analysis, we signify all the measurement materialswith mean±Standard Deviation, all the categorical data with frequency numbers,Hardy-Weinberg test with chi-square test, finally, we use the method of MultifactorDimensionality Reduction to analyze the interactive affect to TCM syndromes ofessential hypertension with the Polymorphism of angiotensinogen and the Polymorphism of angiotensinogen with active substance.Results：1. Comparison of common clinical data between the control group and syndromesof EH group：There is no difference in smoking by comparison of common clinical databetween the control group and syndromes of EH group（P>0.05）,There are differencesin distribution in gender、drinking、age、systolic blood Pressure、diastolic blood Pressureand BMI（P<0.05）,which are comparable.2. Hardy-Weinberg test of distribution of AGT genotype: It comes to a conclusionafter chi-sqrare test that the distribution of genotype in each grop is accord to thehardy-weinberg equilibrium, it goes to show that the Population we study arerepresentative.3. Relevance between syndrome types and AGT active substance: It is the highestin control with AGT active substance,and lowest in the Yin deficiency with yanghyperactivity type,AGT active substance in syndrome types of EH are higher thancontrol group obviously（P<0.05）.AGT active substance among Liver-fire hyperactivitytype、Yin deficiency with yang hyperactivity type、Phlegm-damp retention type、dualDeficiency of yin yang type and control group is statistically significant differently（P<0.05）,among them,Phlegm-damp retention type and control group is lower thenother groups obviously.We found that Liver-fire hyperactivity type、Yin deficiency withyang hyperactivity type、 dual Deficiency of yin yang type and control group isstatistically significant differently（P<0.05）,Phlegm-damp retention type and Deficiencyof yin yang type is statistically significant differently（P<0.05）,Yin deficiency with yanghyperactivity type and Deficiency of yin yang type is statistically significant differently（P<0.05）,other group is not statistically significant differently.4.Comparison of AGT genotype and allele gene among all groups:There issignificant difference in genotype and allele gene AGT rs5049among allgroups(P<0.05),CC genotype is higher in Phlegm-damp retention type,There is no difference in genotype and allele gene in other locus (P>0.05).5. Correlation analysis between SNPs-SNPs ofAGT and syndromes of TranditionalChinese Medicine of EH: rs5049may make a difference in the happening of the dualdeficiency of yin and yang type of EH,the interaction effect among rs2148582、rs4762and rs699may have a association effect in the Yin deficiency with yang hyperactivitytype,the reciprocal action between rs2148582and rs5049may have an effect on thePhlegm-damp retention type.Conclusion:1.There has a influence in EH of people with han nationality in hefei with raise ofAGT active substance,especially in Deficiency of yin yang type;2.There do has a connection between TCM syndromes of EH and Polymorphism ofAGT,There is a significant of AGT rs5049in people with han nationality in hefei;3.rs5049and the dual deficiency of yin and yang type、the interaction effect amongrs2148582、rs4762and rs699and the Yin deficiency with yang hyperactivity type、 thereciprocal action between rs2148582and rs5049and the Phlegm-damp retention typeare related.
