Background:Breast cancer is the most commonly diagnosed cancer and the leading cause of malignancy-related mortality in women worldwide. There is no satisfactory circulating bio-marker currently for early diagnosis, surveillance of relapse or prognosis and therapeutic outcome evaluation. MicroRNA (miRNA) play是a crucial role in oncogenesis and progression of breast cancer. Recent advances in studies of circulating miRNA indicated that it might become a novel stable noninvasive candidate biomarker for breast cancer.Objective:To identify breast cancer related candidate circulating miRNA, and to investigate the correlation between levels of candidate circulating miRNAs and personalized prognostic factors of breast cancer patients, and to evaluate the potential of these miRNAs as candidate bio-markers of breast cancer.Methods:Low molecular weight RNAs including circulating miRNAs were extracted from plasma samples. Levels of three candidate miRNAs were amplified by real-time quantitative polymerase chain reaction (RT-qPCR) in plasma of43breast cancer patients and11breast fibroadenoma patients as controls. The correlation between levels of circulating miRNAs and personalized prognostic factors were investigated, including clinical stage, molecular subtype, receptor status, LN metastasis, grade, peri-vascular invasion (PVI), etc.Results:we found that the level of let-7f was significantly elevated in breast cancer with lymph node metastasis (p=0.033) and high grade breast cancer (p=0.011). The level of miR-125a-5p was decreased in breast cancer with PVI (p=0.004).Conclusions:The level of plasma let-7f may correlate with lymph node metastasis and histological grade of breast cancer. The level of plasma miR-125a-5p may correlate with breast cancer peri-vascular invasion. Our pilot study suggested that circulating miRNA might have correlations with personalized prognostic factors of breast cancer and become candidate breast cancer biomarker.
