Intracerebral hemorrhage(ICH) also called non-wounded cerebral hemorrhage, is a common cerebrovascular diseases and one of the main threats of aging’s healthy. For the treatment of cerebral hemorrhage, Chinese medicine has an unique effect.Polydatin is one of the main active ingredient of traditional Chinese medicine polygonum which has promoting blood circulation activities and removing blood stasis. Modern pharmacological studies have demonstrated that polydatin has multiple biological activities such as improve blood rheology, anti-thrombus formation, anti-oxidation, regulate nitric oxide, affecting ion channels and protein kinase. This study is to investigate the effect and mechanisms of polydatin on brain injury after ICH hemorrhagic induced by collegenase in rats and neuronal injury induced by thrombin in cultured neurons.1. The effect and mechanisms of polydatin on brain injury after ICH in rats induced by collgenaseThis section was intended to investigate the effect and mechanisms of polydatin on brain injury after ICH in rats. The intracerebral hemorrhage model in rats was induced by local caudate nucleus injection of type VII(0.25u/μL) collagenase and were treated with polydatin100,50,25mg/kg(2times/day,3days). Neunological score, the cerebral index, the content of water, red blood cell aggregation, red blood cell deformability and brain tissue HE staining were tested at the third day after the operation. The activity of superoxide dismutase(SOD), the content of malondialdehyde(MDA) in serum and brain tissue were measured. The expression of matrix metalloproteinase-9(MMP-9), aquaporine-4(AQP-4), cell apoptisis and apoptisis associated protein Bcl-2, Bax around hemotoma were detected by immunohistochemistry and western-blot. The results suggested:1.1Polydatin has the protective effect on the brain injury in rats induced by ICH. Compared with the ICH model group, polydatin(25mg/kg) could significantly reduce the nerve symtoms(P<0.05). Polydatin could attenuate the cerebral index, but there is no significant difference. Polydatin(25mg/kg) could significantly reduce the content of water in ICH model rats brain tissue (P<0.05). polydatin(100,50,25mg/kg) could significantly reduce the whole blood aggregation in ICH model rats(P<0.01or P<0.05), polydatin(100mg/kg) can significantly improve the erythrocyte deformability in ICH model rats (P<0.05). HE staining results showed that polydatin (100,50,25mg/kg) could reduce the bleeding area, repair the hematoma surrounding tissue, reduce the swelling of nerve cells. The effect of polydatin(100mg/kg) was more significant.1.2Polydatin has a certain antioxidative effect. Compared with the cerebral hemorrhage model group, polydatin (100,50,25mg/kg) could markedly increase the activity of SOD, reduce the content of MDA in serum(P<0.01or P<0.05); polydatin(100,50,25mg/kg) could increase the activity of SOD, reduce the content of MDA in brain tissue, but there is no significant difference.1.3Polydatin has a certain improving cerebral edema effect. The immunohistochemistry staining results that polydatin(100,50,25mg/kg) could obviously reduce the expression of MMP-9and AQP-4(P<0.01or P<0.05) in the brain tissue around herotoma after ICH. Western-blot assay results showed that polydatin(100,50,25mg/kg) could decrease the expression of MMP-9and AQP-4, but there is no significant difference.1.4Polydatin has a certain against the neuronal apoptosis effect. The immunohistochemistry staining results that polydatin(100,50,25mg/kg) could distinctly reduce the TUNEL staining positive cells、the expression of Bax and the Bax/Bcl-2value and increase the expression of Bcl-2(P<0.01or P<0.05). Western-blot assay show that polydatin(100,50,25mg/kg) could decrease the expression of Bax, increase the expression of Bcl-2, but there is no significant difference.It was indicated that polydatin has the protective effect on the brain injury in rats induced by ICH, which maybe related with antioxidative effect, improving cerebral edema and against the neuronal apoptosis.2. The effect of polydatin on neurons injury induced by thrombinThis section was intended to observe the effect of polydatin on neurons injury induced by thrombin. Cortical neurons derived from the pregnant16-17d SD rat were cultured for6days, thrombin (150U/mL) and polydatin (15.625,31.25,62.5μmol/L) were added into the culteure medicum and then incubated for3days. The cell viability was determined by MTT method. The results showed that the cell viability of polydatin(31.25μmol/L) was84.67%, is higher than the model group (P<0.01). It was indicated that polydatin has protective effect on thrombin-induced neuronal injury.In summary, polydatin has protective effect on intracerebral hemorrhage injury in rats, which maybe related with reducting the free radical damage, attnusating the brain edema, inhibiting neuronal apoptosis and protecting neurons.
