Objective: The present study is to determine the effects of Shenqi compound recipe on the macroangiopathy in spontaneously diabetic KKAy mice via a split-side researchMethods:seventy-five KKAy male mice(twenty-week-old) whose blood glucose is greater than13.9mmol/L were randomly divided into five groups:KKAy group (normal saline10ml·kg-1cd-1), diabetic macrovascular disease group (0.2mg·ml-1·d-1L-NAME in drinking water with high fat diet), Shenqi Compound of Recipe group (0.2mg·ml-1·d-1L-NAME in drinking water plus Shenqi Compound of Recipe14.4g·kg-1·d-1by intubation with high fat diet), Yiqiyangyin group (0.2mg·ml-1·d-1L-NAME in drinking water plus11.7g·kg-1·d-1Yiqiyangyin compound by intubation with high fat diet), huoxue group (0.2mg·ml-1·d-1L-NAME in drinking water plus2.7g· kg-1·d-1huoxue Compound by intubation with high fat diet). C57BL/6J mice (normal saline10ml.kg-1.d-1) had been used as control group. During the experiment, the food and water intake had been recorded everyday and the blood glucose concentration had been investigated every week. After8weeks, all mice had been sacrificed. The blood samples had been collected for serum lipid (total cholesterol, TC; triglyceride, TG; high density lipid, HDL; low density, LDL; insulin, INS) measurements. The vascular of aorta abdominalis had been removed for HE stain. The thoracic aorta had been collected for GO biological procession analysis by genechip for gene annotation in function and pathwayResults:the inner membrane of the vascular of aorta abdominalis has been observed that higher swollen and continuous exfoliation in diabetic macrovascular disease group than that in other groups by HE stain. In addition, the concentrations of blood glucose, serum lipid and INS, and HOMA-IR were significant increased in diabetic macrovascular disease group than that in other groups. The concentrations of blood glucose, serum lipid, and insulin resistance were significant decreased in Shenqi Compound of Recipe group, Yiqiyangyin group and huoxue group than that in diabetic macrovascular disease group. The inner membrane of the vascular of aorta abdominalis has less swollen and continuous exfoliation in the Shenqi Compound of Recipe group than that in diabetic macrovascular disease group. Moreover, the insulin resistance and the serum concentration of TG and LDL were tending to lower in Shenqi Compound of Recipe group than that in Yiqiyangyin group and huoxue group. Furthermore, the blood glucose concentration was more stable in Shenqi Compound of Recipe group than that in Yiqiyangyin group and huoxue group. In genechip analysis, the Shenqi Compound of Recipe could decrease the diabetic macroangiopathy via several mechanisms including cell apoptosis, cell proliferationConclusion::The Shenqi Compound of Recipe can significantly decrease insulin resistance, and the concentrations of blood glucose and serum lipid. Moreover, the inner membrane of the vascular of aorta abdominalis has been less histological damaged changed after the Shenqi Compound of Recipe treatment which might be related to several mechanisms.These results revealed that the beneficial effects of Shenqi Compound Recipe on the macroangiopathy might be better than that in split-side formulas in spontaneously diabetic KKAy mice which supported that the combination of the Shenqi Compound of Recipe is rationality and scientificalness...
