| Angelica dahurica Radix (Chinese name:Baizhi), which is one of the famous traditional Chinese medicines,consists ofthe dried roots of Angelica dahurica(Fischex Hoffm.) Benth.et Hook.f or Angelica dahurica (Fisch ex Hoffm.) Benth.et Hook.f.var.formosana (Boiss) Shan et Yuan of Umbelliferae. This herbal medicine has also been claimed to be effective in the treatments of acne, erythema, headache, toothache, sinusitis, colds and flu.In the present study, a sensitive method using off-line2D-HPLC-ESI/MS" to identify coumarins from A. dahuricawas developed for the first time. Based on analysis of ESI/MS" behaviours of these coumarins, possible fragmentations and mechanisms were reconsidered. As a result,50coumarins, including35coumarins and15dimers of linear furanocoumrins (LFCs), were found from A. dahurica. Most of the coumarins with trace amounts are dimers of LFCs. Purification of these structurally diverse dimers would be meaningful, and using HPLC-ESI/MSn guided separation would increase the efficiency of separating these structurally diverse coumarins.Ditaohuaconsists ofthe dried aerial parts of Urena lobata L.(Malvaceae). As a traditional Chinese medicine, it has been used to treat cold, fever, pain or numbness caused by rheumatism, dysentery, oedema, gonorrhea, leucorrhea, haematemesis, carbuncle, trauma, bleeding ect.Ditaohua has not yet been listed in the Chinese Pharmacopoeia, however,some herbal medicine preparations that containDitaohua,have been archived in Chinese Pharmacopoeia, such asHuahong tablets. In some folk standards, there are only some simple rules about source and processing of this herbal medicine,which is far from meeting the requirements of the treatment standardsIn current study, we developed a fullquality standard of Ditaohua. Its fiber traits were characterized using microscopy, and then thin layer chromatography was used to identify the crude material. The moisture, ash and extractives were also studied. At last the determination of tiliroside, which is the major component in Ditaohua, was also performed. Above all, a comprehensive study of Ditaohuawasachieved and the data was true and reliable. This study laid a solid foundation for the addition of Ditaohua in Chinese Pharmacopoeia (2015edition) and provided strong basis for its regulated use in clinical and safety. |
