DNA barcoding technology is the latest development in the field of molecular biology techniques of molecular identification, i.e. by using a standard with sufficient variability, and relatively easy to amplify short DNA fragment to identify specific species and the species diversity and simultaneously to achieve fast, accurate identification and authentication of species. And it is the research hotspots of biological classification and identification in recent years. To attribute the revision and improvement of quality standards Acorus Tatarinowii, this study focused on the DNA barcode technology for the analysis of Acorus medicinal plants, as well as feasibility of identification of the Acorus Tatarinowii and its analogues adulterants(Acorus Calamu,Acorus Gramineus,A.tatarinowii sehott var.flavo-marginatus K.M.Liu and Acorus Macrospadiceus), establishment of the PCR reaction conditions for molecular identification of Acorus Tatarinowii, and further investigation of the genus Acorus species ascription. The study includes:I The collected Acorus samples were identified as Acorus Tatarinowii or its analogues adulterants(Acorus Calamu,Acorus Gramineus,A.tatarinowii sehott var. flavo-marginatus K.M.Liu,Acorus Macrospadiceus) by pharmacognostic method. Ⅱ These identified samples were studied by the DNA barcoding technology1. Screening the best fit DNA barcoding for the genus AcorusFour candidate DNA barcode sequence ITS2, psbA-trnH, matK, rbcL of the genus Acorus were amplified and sequenced. By studing the intraspecific variation interspecific variation and species identification capability, the results indicated that the ability of ITS2to identify the genus of Acorus is the best, which can be used as DNA barcoding.2. The specificity PCR identification of the Acorus Tatarinowii.By primer design software designed specific primer for identification of Acorus Tatarinowii:ScpuJbie Upper and ScpuJbie Lower, and studied the optimal annealing temperature. The results showred that when the annealing temperature was72℃, this specific PCR can effectively distinguish the Acorus Calamu and Acorus Tatarinowii.Ⅲ Investigation of the botanical origin of Acorus base on DNA barcodeing.Based on acquired ITS2sequences of the genus Acorus, combined with the corresponding DNA barcode sequences from Genbank database, bioinformatics analysis showed the phylogenetic and taxonomic relationships between the Acorus and Araceae and support the view raised by Flora of China that the Acorus should be separation alone, became a subject of an independent, which provided a molecular basis for the original species of Acorus and a scientific basis for the botanical origin of Acorus at the genetic level.
