Objective To investigate the relationship between postoperative cognitive dysfunction (POCD) of aging model rats and stress. Furthermore, to observe the therapeutic effect of electroacupuncture on rats suffered from POCD and the regulatory mechanisms of stress and immune。Methods One hundred famale SD rats (12month old) were randomly divided into control group (group C, n=10), model group(group M, n=30),electroacupuncture group (group E, n=30) and Drug group (group D, n=30). According to the execution time after the POCD model established, the model group was divided into model1days group (group Mid, n=10), model3days group (group M3d, n=10)and model7days group (group M7d, n=10); the electroacupuncture group was divided into electroacupuncture1days group (group Eld, n=10), electroacupuncture,3days group (group E3d, n=10) and electroacupuncture7days group (group E7d, n=10); the drug group was divided into Drug1days group (group D1d, n=10), Drug3days group (group D3d, n=10) and Drug7days group (group D7d, n=10). Train the rats in morris water maze once a day for five consecutive days.Afer training,all the groups were adopted the model of partial hepatectomy except group C. The group E received electroacupuncture and the group D received intraperitoneal injection of10%minocycline (40mg/kg) after2hours of postoperation,once a day.Group C,group3days and group7days got morris water maze experiment before the execution. Experiment indicators:(1) Detected cognitive and memory function by morris water maze;(2) Measured the levels of ACTH, CORT and CRH in blood serum by enzyme linked immunoadsorbent assay (ELISA);(3) Detected GR, MHC II, CD200, CD200R expression in hippocampus by immunohistochemistry.(4) Detected mRNA Expression of GR, TLR2, TLR4, MHC II by Fluorescence quantitative PCRResults (1) In the training time before surgery,there was no significant difference about learning and space exploration ability in10groups. Between any two groups,the difference of costing time finding the platform was not significant(P>0.05). Compared with group C,target quadrant residence time of group M3d and group M7d was significantly lower in the water maze test before execution (P<0.01). Compared with group M3d, target quadrant residence time of group E3d and group D3d was significantly longer in the water maze test before execution (P<0.05). Compared with group M7d, target quadrant residence time of group E7d and group D7d was significantly longer in the water maze test before execution (P<0.05).(2) Compared with group C, the levels of ACTH, CORT and CRH increased significantly (P<0.05) in group M1d, group M3d and group M7d. Compared with group M1d, the levels of ACTH decreased significantly (P<0.05) in group D1d. Compared with group M1d, the levels of CORT decreased significantly (P<0.05) in group E1d and group D1d. Compared with group M3d, the levels of ACTH, CORT and CRH decreased significantly (P<0.05) in group E3d and group D3d. Compared with group M7d, the levels of ACTH, CORT and CRH decreased significantly (P<0.05) in group E7d and group D7d.(3) mRNA Expression of GR of group M1d, group M3d and group M7d was decreased significantly than that of group C(P<0.05). mRNA Expression of GR of group Eld, group D1d was increased more significantly than that of group Mld(P<0.05). mRNA Expression of GR of group E3d, group D3d was increased more significantly than that of group M3d(P<0.05). mRNA Expression of GR of group E7d, group D7d was increased more significantly than that of group M7d(P<0.05).mRNA Expression of GR of group was increased more significantly than that of the sham operation group(P<0.05). mRNA Expression of TLR2/4of group M1d, group M3d and group M7d was decreased more significantly than that of group C(P<0.05). mRNA Expression of TLR2/4of group E3d, group D3d was increased more significantly than that of M3d(P<0.05). mRNA Expression of TLR2/4of group E7d, group D7d was increased more significantly than that of M7d(P<0.05). mRNA Expression of MHC II of group M1d, group M3d and group M7d was decreased more significantly than that of group C(P<0.05). mRNA Expression of MHC II of group E3d, group D3d was increased more significantly than that of M3d(P<0.05). mRNA Expression of MHC II of group E7d, group D7d was increased more significantly than that of M7d(P<0.05).(4) The number of GR positive cells of group M1d, group M3d and group M7d were decreased significantly as compared with group C(P<0.05). The number of GR positive cells of group Eld, group D1d were increased significantly as compared with group M1d (P<0.05). The number of GR positive cells of group E3d, group D3d were increased significantly as compared with group M3d (P<0.05). The number of GR positive cells of group E7d, group D7d were increased significantly as compared with group M7d (P<0.05). The number of MHC II positive cells of group M1d, group M3d and group M7d were increased significantly as compared with group C(P<0.05). The number of MHC II positive cells of group E3d, group D3d were decreased significantly as compared with group M3d (P<0.05). The number of MHC II positive cells of group E7d, group D7d were decreased significantly as compared with group M7d (P<0.05). The number of CD200positive cells of group M1d, group M3d and group M7d were increased significantly as compared with group C(P<0.05). The number of CD200positive cells of group Eld, group D1d were decreased significantly as compared with group M1d (P<0.05).The number of CD200positive cells of group E3d, group D3d were decreased significantly as compared with group M3d (P<0.05). The number of CD200positive cells of group E7d, group D7d were decreased significantly as compared with group M7d (P<0.05). The number of CD200R positive cells of group M1d and group M7d were increased as compared with group C.The difference of CD200R positive cells was not significant(P>0.05). The number of MHC II positive cells of group M3d was increased significantly as compared with group C(P<0.05). The number of MHC II positive cells of group D3d, group D7d was decreased significantly as compared with group M3d and group M7d(P<0.05). The number of CD200R positive cells of group E1d, group D1d were decreased as compared with group M1d. The difference of CD200R positive cells was not significant(P>0.05). The number of CD200R positive cells of group E3d was decreased as compared with group M3d. The difference of CD200R positive cells was not significant(P>0.05).The number of CD200R positive cells of group E7d was decreased as compared with group M7d. The difference of CD200R positive cells was not significant(P>0.05).Conclusion Electroacupuncture can achieve the aim of improving POCD by adjusting hypothalamus-pituitary-adrenal axis (HP A axis) of POCD model rat, improving sustained damage to the hippocampus from increasing stress hormone, reducing the degree of microglia activation and inducing hippocampal inflammationthe.proinflammatory.
