Secreted Frizzled-related Protein Family Gene Expression Analysis In Patients With Cumulus Granulosa Cells In Women With Polycystic Ovary Syndrome

Objective:To investigate the gene expression profiles of sFRP family members in cumulus cells of oocytes at different mature stages in PCOS or non-PCOS patients.Methods:After controlled ovarian stimulation (COS), a total of197cumulus cell samples (CCPCOSs=97, CCnon-PCOS=100) were obtained from27patients (PCOS=11, non-PCOS=16) during IVF cycles. All the CC samples were divided into four groups:CCN-MⅡ, CCN-MI+GV, CCP-MⅡ, CCP-MI+GV based on the maturation of oocyte. Quantitative real time-PCR (qRT-PCR) was performed in this study.Results:1.Only the expression of sFRP4was (7.80±1.21)-fold(p<0.05) higer in CCP-MⅠ+GV than CCP-MⅡ of PCOS patients.2. sFRP4and sFRP5had higher expression level (p<0.05) in CCP-MⅠ+GV than CCP-MⅡ of non-PCOS patients, i.e., the expression level of sFRP4was (7.04±1.30)-fold in CCP-MⅠ+GV than CCP-MⅡ, the expression level of sFRP5was (2.84±0.73)-fold in CCP-MⅠ+GV than CCP-MⅡ.3. There was no significant difference in expression of sFRP4between PCOS and non-PCOS patients.Conclusion:These observations demonstrated the expression level of sFRP4was decreased as the follicles growing. However, the expression of sFRP5merely came up in cumulus cells from non-PCOS patients.
