In the traditional nitrogen cycle system, usually, ammonia oxidized byammonia oxidizing bacteria, from ammonia to nitrite, then into nitrates, thebiological nitrogen cycle was beginning with this. In recent years, the discoveryof Anaerobic ammonia Oxidation (anammox) and anammox bacteria, with thedeepening research, changed people’s understanding of the traditional nitrogencycle system. This paper aims to use molecular biology technique such asPCR(polymerase chain reaction) technology, clone libraries, to validate that theanaerobic ammonia oxidation bacteria can live in different natural wetlandenvironment (sediments), and the existence under the artificial environment(activated sludge), we also analyzed the biological diversity of different samples.It is obtained the positive results of anammox bacteria16S rRNA gene by PCRtechnology, and determined the abundance of anammox bacteria by thequantitative polymerase chain reaction (qPCR), thus proving the universaldistribution of anammox bacteria. Then building the clone library and anammox16S rRNA sequence phylogenetic tree by the sample of sediments and activatedsludge, made analysis about the biological diversity, found that the biologicaldiversity of samples in the north is more than in the south. The cloning sequencefrom the sample of the natural wetland sediments mostly showed high similaritywith Brocadia and Kuenenia; and the cloning sequence of activated sludge, madea higher biological diversity than the sediment samples’, showed a highsimilarity with Brocadia, Scalindua and Jettenia. From the sequence phylogenetictree and distribution of the biological diversity, the addition of anammox bacteriainto the artificial environment just like anaerobic reactor has a high feasibility.
