| The research on the active of single factor of the natural plant is more,but the research on the different factors from the same source have no reports.therefore,the studies take the xinzheng huizao as the raw materials.By using the method of in vitro antioxidant capacity and in vivo antioxidant capacity to explore the cooperative antioxidant activity of Jujube polysaccharide and flavonoid and speculate the reasons of interaction.The experiments mainly reached following results:1ã€Using the method of in vitro antioxidant capacity to explore the cooperative antioxidant activity of Jujube polysaccharide and flavonoid(1)The research focus on the ability of scavenging the ·OH free radicals in the experimental composition range of 0.1,0.3,0.5,1.0,1.5,2.0mg/m L.When the solution concentration is less than 1 mg/m L,the ability of scavenging the ·OH free radicals of JP, JF and JP + JF is:JF>JP+JF>JP,the differences is significant.(P<0.05).When the solution concentration is more than 1 mg/m L,the ability of scavenging the ·OH free radicals of JP, JF and JP+JF is:JP+JF>JF>JP,the differences is significant.(P<0.05).(2)The research focus on the ability of scavenging the DPPH· free radicals in the experimental composition range of 0.1,0.3,0.5,0.7,0.9,1.0mg/m L.The ability of scavenging the DPPH· free radicals of JP, JF and JP+JF is:JF>JP+JF>JP,the differences is significant.(P<0.05).(3)The research focus on the ability of scavenging the ABTS+· free radicals in the experimental composition range of 0.03,0.07,0.10,0.15,0.20,0.30mg/m L.When the solution concentration is less than 0.1 mg/m L,the ability of scavenging the ABTS+· free radicals of JP, JF and JP+JF is:JF>JP+JF>JP,the differences is not significant.(P>0.05).When the solution concentration is more than 0.1 mg/m L,the ability of scavenging the ABTS+· free radicals of JP, JF and JP + JF is:JP+JF>JF>JP,the differences is also not significant.(P>0.05).(4)The research focus on the ability of scavenging the total antioxidant activity(FRAP value) in the experimental composition range of 0.1,0.3,0.5,1.0,1.5,2.0mg/m L.The FRAP value of JP, JF and JP+JF is:JP+JF>JF>JP,but when the concentration is lesson than 1.5 mg/m L,the differences is not significant.(P>0.05).When the concentration is more than 1.5 mg/m L,the differences is significant.(P<0.05).(5)Jujube polysaccharide and flavonoid had the positively cooperative antioxidant activity on the four reaction system. The cooperative antioxidant activity of antioxidants had the positively relation to the effective concentration of antioxidants and the antioxidant activity of antioxidants themselves(R2>0.82;P<0.01),was also related to the reaction systems.(6)As a representative selection of ·OH reaction system,the best ratio of cooperative antioxidant activity of Jujube polysaccharide and flavonoid was 1:1,the concentration was 1.5mg/m L,the ability of scavenging free radicals was 97.88%,it was 63.33% higher than Jujube polysaccharide,16.36% higher than Jujube flavonoids.2ã€Using the method of in vivo antioxidant capacity to explore the cooperative antioxidant activity of Jujube polysaccharide and flavonoid(1)Compared with the normal control group, the spleen index and thymus index of model group decreased significantly(P<0.01,P<0.05), it suggests that the model is succesful.The ability of improving the spleen index and thymus index of JP, JF and JP+JF middle dose is:JP+JF>JF>JP,the differences is not significant(P>0.05).(2)The ability of improving the activity of SOD enzyme of JP, JF and JP+JF middle dose is:JP+JF>JF>JP,the differences of JF>JP is significant(P<0.01),the differences of JP+JF>JF is not significant(P>0.05).(3)The ability of improving the activity of GSH-PX enzyme of JP, JF and JP+JF middle dose is:JP+JF>JF>JP,the differences of JF>JP is not significant(P>0.05),the differences of JP+JF>JF is significant(P<0.05).(4)The ability of improving the content of HYP of JP, JF and JP+JF middle dose is:JP+JF>JF>JP,the differences is significant.(P<0.05).(5)The ability of reducing the content of MDA of JP, JF and JP+JF middle dose is:JP+JF>JF>JP,the differences is significant.(P<0.05).(6)The ability of improving the content of brain neurotransmitter of JP, JF and JP+JF middle dose is:JP+JF>JF>JP,the differences is significant.(P<0.05).(7)The ability of improving the content of serum neurotransmitter of JP, JF and JP+JF middle dose is:JP+JF>JF>JP,the differences is significant.(P<0.05).(8)JP and JF presents the cooperative antioxidant activity in aging mice.The thymus index and spleen index has significant positive correlation(R2 = 0.85,P < 0.01).The activity of SOD enzyme and GSH-PX enzyme and the content of MDA between the different tissue has significant positive correlation(R2>0.99,P<0.01;R2>0.98;P<0.01;R2>0.95;P<0.01).The content of MDA in the skin and serum has significant positive correlation(R2=0.99; P<0.01).The pats of neurotransmitter in brain and serum has significant positive correlation(R2>0.74,P<0.05;R2>0.76, P<0.05).(9)When JP and JF role in mice.It is not applied to a tissue or organ, but to the whole body system. |
