Hemoglobin protein has a unique features that can bind and transport oxygen to various organs.Therefore,separation and purification of hemoglobin is very imp-ortant. At present,various methods such as centrifugation,crystallization,adsorption,chroma-tography,ultrafiltration and precipitation have been used to isolate hemoglobin from whole blood.Although hemoglobin with high purity could been obtained by these methods,but large scale production is difficult,sometimes ultracentrifuge or filter specific chromatograp-hy media or other expensive facilities are needed.For these reasons, molecular imprinting technology were employed to achive selective recognition of hemoglobin.Magnetic protein molecular imprinted material were prepared and used to recognize hemoglobin.In this research,magnetic molecularly imprinted materials based onbovine hemoglobin protein were obtained,and its recognition performance were investigated.First,Fe3O4 particles were synthesized by hydrothermal method and coated with SiO2 on the surface. Throughgraft modificationon particles surface.In order to introduceamino onto magnetic material surface, the particles were further modified with mPDA-IPTS.The template protein was bond to the surface of magnetic materials loaded with Cu+based on the interaction between copper ions andbovine hemoglobin. Subsequently, magnetic molecularly imprinted polymers were prepared by using mPDA as functional monomer and ammonium persulfate as initiator.The obtained materials were characterized by IR,SEM,TEM,and TGA,respectively. The recognition property of magnetic materials were evaluated. The results indicated that the magnetic imprinted materials showed highly selectivity and binding capacity to the template protein, the adsorption capacity of bovine hemoglobin material reached 389mg/g, adsorption could be completed within 3h. The resulted materials were also used toselectively recognize target proteins from complex samples. Imprinted material produced by this method is expected to fabricate imprinted materials for other proteins. |
