Red jujube is buckthorn plant genus ripe fruit, rich in amino acids, polysaccharides, saponin, polyphenolic compounds, some trace elements required by the human body and other biologically active ingredients. With a unique nutritional value, it can lower cholesterol levels, nourish the stomach and prevent arteriosclerosis.Red Pigment can be extracted from the jujube skin, a kind of byproduct during the further processing of the jujube, and it can serve as a natural food coloring which not only provides foods with a pleasing color but also stimulate the appetite of consumers, making itself a new member to the grand family of edible natural pigment. Also, it succeeds in enhancing added values and boosting economic benefits. Red jujube skin extracted from the bark of jujube and detailed qualitative and quantitative analysis of effective components Red jujube pigment may be contained in the establishment of mouse renal oxidative stress-induced injury model to study KBrO3 Red Pigment oxidative stress Protective effects of kidney injury caused by high performance liquid chromatography polyphenols Red pigment contained components were analyzed to provide a theoretical and practical basis for the study of Red pigment structures. The main results are as follows:1.Establish KBrO3 mice models to induce oxidative stress kidney damage. Mice were injected intraperitoneally with 200mg/kg of KBrO3 in order to induce oxidative stress kidney damage, respectively 3,6,9,12,18,24h after sacrifice. Later, the contents of mouse serum Cr, BUNmouse kidney tissue MDA, NO content and GSH-Px activity model induced renal oxidative stress KBrO3 injury is detected. Experimental results show that:Intraperitoneal injection of KBrO3 can induce renal injury in mice, especially injury 6h for the best effect, when serum BUN and Cr content at this time significantly increased in mice and kidney tissue MDA and NO also rise significantly (P<0.05), whereas kidney vitality of tissue GSH-PX decreased dramatically. Description With the establishment of effective potassium bromate oxidative stress renal injury model mice (P<0.05).2.The study of stress kidney damage in mice oxide Red Pigment in terms of KBrO3 reaction. The mice were randomly divided into control group, model group, positive control group (VC) and Red Pigment low-dose group (500 mg/kg), red Red jujube pigment dose group (1000 mg/ kg), red pigment Red jujube high-dose group (2000 mg/kg), After ten days of feeding, the mice are injected with KBrO3, and the serum and kidney tissue content of oxidation products of Cr, BUN, MDA, NO, GSH and antioxidant enzymes SOD, GSH-PX activity and T-AOC levels and organ coefficient are subsequently detected. The results reveal:Red Pigment can effectively reduce Cr, BUN, MDA, NO, GSH content in mice oxidation products, while reducing antioxidant enzymes SOD, GSH-PX activity and T-AOC levels and organ coefficient in mice the degree of renal damage. It reveals that, Red Pigment, by scavenging animals accumulated body of free radicals, can improve its high level of oxidative stress, oxidative stress in mice renal injury, which proves to have a good protective effect.3.To achieve a qualitative and quantitative analysis of Red jujube powder, extracted pigment and purified polyphenols pigment through the use of high-performance liquid chromatograph and ferulic acid, rutin, quercetin, caffeic acid, kaempferol and coumaric acid as an index component for Red jujube powder after pigment extracted and purified polyphenols pigment to determine the content of the active ingredient. Chromatography conditions:C18 column, methanol mobile phase A, B acetic acid-water (1:250v/v) solution; the total flow rate of 1.0 mL/min, column temperature 30℃, the injection volume 20μL, detection wavelength 290nm. The results show that:after extraction and purification steps, the most abundant polyphenols in Red jujube powder turn out to be kaempferol and rutin comes second, which is followed by kaempferol; and the content of most of the polyphenols has undergone a very significant increase.
