Most liquid preparation of Chinese Medicine are made from Chinese herbal compound prescription. A variety of mixture, extraction, concentration, and filtration, will not only change the physical properties of herbal medicine, it can also change their chemical composition, respectively.The stability and security of liquid preparation of Chinese Medicine has always been a difficult research in Chinese Medicine. Modern scholars recognized that quality of preparation of Chinese medicine is no longer to check the quality of the product, but also the real-time monitoring for the process of produtive preparation, to control the productive process and ensure stability and reliability of product quality in each of links. Nowadays state-level to control the quality of Chinese Medicine and Chinese preparations is qualitative identification and content determination.UV, TLC and HPLC are common measurements. This topic is on the basis of scholars in Chinese Medical to discuss the feasibility and practicability of determination by NIR technology and to provide reference for the current quality evaluation system. Apply the near-infrared spectral technology into the process analysis, not only achieve the purpose of real-time monitoring, but also ensure that the detection process is green, fast, simple and convenient.(1) Research on the quantitative model of Yinhuang Oral Liquid by Near-infrared analysis, not only can achieve real-time monitoring aim, but also ensure the testing process to be green, fast and simple.Objective:This research is to study and develop a method for online monitoring of the industrial production of active components that are Chlorogenic acid and Baiclin of Yinhuang oral liquid by using near-infrared(NIR) spectral technology.The chemical value of the Chlorogenic acid and Baiclin were analyzed by the high performance liquid chromatography(HPLC). Methods:After collecting different batches of Yinhuang oral liquid, we detected their NIR spectra. The quantitative analysis models of the above two compounds were established using partial least square(PLS).During modeling, coefficient of determination(R2), root mean square errors of cross-validation(RMSECV) and ratio of performance to standard deviate(RPD) were regarded as the indexes to select optimal wave numbers and preprocessing methods.Results:The optimal wave numbers of Chlorogenic acid and Baiclin were both 9401.6~7826 cm-1; R2 were 0.9852,0.9881, respectively; RMSECV were 0.00296,0.018, respectively; RPD were 8.23,9.27, respectively.The above developed model was used for unknown contents of Chlorogenic acid and Baiclin of Yinhuang oral liquid.The value of Mash dist of predicted results were less than 1, the relative errors were 2.12%,1.67%, respectively.Conclusion:This method was proved to be fast, convenient and precise.It can be used for online assaying and quality control of Chinese drugs pharmaceutics in manufacture.(2) Establishment of the Model for Online Monitoring by Near-infrared Spectoroscopy of Yinzhihuang Oral Liquid in Extraction ProcessObjective:This research is to investigate the effective components dissolution using near-infrared(NIR) spectroscopy for the on-line monitoring of Chlorogenic acid, Baiclin and Geniposide in the extracting process of Yinzhihuang. Methods:After collecting Yinzhihuang extract at different points in time, we detected their NIR spectra. The chemical value of the Chlorogenic acid, Baiclin and Geniposide were analyzed by the high performance liquid chromatography(HPLC). The quantitative analysis models of the above three compounds were established using partial least square(PLS).During modeling, coefficient of determination(R2), root mean square errors of cross-validation(RMSECV) and Rank were regarded as the indexes to select optimal wave numbers and preprocessing methods.Results:The optimal wave numbers of Chlorogenic acid, Baiclin and Geniposide were 9401.6~7498.2 cm-1, 9401.6~8448.9 cm-1,9401.6~7498.2 cm-1, respectively; R2 were 0.8873,0.9072, 0.9538, respectively; RMSECV were 0.119,0.372,0.0306, respectively; Rank were 6,3,6, respectively.The above developed model was used for unknown contents of Chlorogenic acid, Baiclin and Geniposide of Yinzhihuang oral liquid in the parallel condition. The value of Mash dist of predicted results were less than 1, the average relative errors were 2.14%,0.47%,2.05%, respectively.Conclusion:This method was proved to be fast, convenient and precise.Providing a scientific basis for process control of Chinese medicine.(3) Research on the content determination of San’ao Tang in Extraction Process by Near-infrared SpectoroscopyObjective:This research is to assay the main components by using near-infrared(NIR) spectroscopy for the real-time monitoring of Ephedrine(in the form of Ephedrine hydrochloride), Pseudoephedrine(in the form of Pseudoephedrine hydrochloride) and Amygdalin in extraction of San’ao Tang.Methods:Collected extraction of San’ao Tang in every minute, we detected their NIR spectra. The chemical value of the Ephedrine, Pseudoephedrine and Amygdalin were analyzed by the high performance liquid chromatography(HPLC).The quantitative analysis models of the above three compounds were established using partial least square(PLS).During modeling, coefficient of determination(R2), root mean square errors of cross-validation(RMSECV) and Rank were regarded as the indexes to select optimal wave numbers and preprocessing methods.Results:The optimal wave numbers of Ephedrine, Pseudoephedrine and Amygdalin were 9401.6~7498.2 cm-1; R2 were 0.9349,0.9274,0.8557, respectively; RMSECV were 0.00972,0.00819,0.0532, respectively; Rank were 8,8,4, respectively.The above developed model was used for detecting Ephedrine, Pseudoephedrine and Amygdalin in another batch of San’ao Tang.The value of Mash dist of predicted results were less than 1, the average relative errors were 2.41%,2.74%,2.56%, respectively.Conclusion:This method was proved to be good predictive performance.Providing a scientific basis for process control of Chinese medicine. Both can be measured in real-time and indicate the end of the extraction.
