Copper resources reserves in Yunnan ranks second in China. It is expected that it has huge reserves in the future. In recent years, Yunnan has been proven a number of large deposits. The climate of Yunnan is mild and moist, which is favorable for the application of the bio metallurgy technology. Therefore, the study of the biological leaching of copper ore is of great significance to the development and utilization of the resources of Yunnan copper mine and the copper industry in the whole country.In this paper, the basic research on the bio leaching process of copper sulphide ore including the thermodynamic calculation of the mineral’s bio metallurgical leaching process. The results provided the thermodynamic basis for the process of bio metallurgy that the predominance area of Cu2+ in the growth of bacteria was that the pH value is1.0-2.5 and the redox potential is 400-900 mV. The acid mine drainage collected from Damai copper mine in Yunnan, Wulonggou gold mine in Qinghai and Zijinshan copper mine in Fujian using 9K liquid medium for screening culture. The strains were mixed and domesticated to obtain higher activity. And try to screen and culture Moderately thermophilic bacteria and Acidiphilium cryptum G30-1 strains. The pyrite and chalcopyrite two minerals were cut, polished and mosaic, by scanning electron microscopy were detected to observe the chalcopyrite and pyrite mineral surface erosion and bacterial adsorption.The two kinds of Damai copper ore Yunnan Province were studied by flask andcolumn leaching experiment. The results showed that The Cu extraction of Fuyuan copper mine is above 90%. Through laboratory column leaching of two copper sulfide ore, leaching property, acid consumption and leaching rate of process parameters have been investigated. The results showed that the acid consumption of ore was low, and easy to leaching. The DM-1 ore is partially oxidized on shallow layer of sulfide ore, The dissolution rate of Cu in ore is relatively fast. After 20 days, the Cu extraction of DM-2ore is about 62.51%, while the Fe extraction is about 36.14%. It shows that the Damai copper sulfide ore is easy to deposit, and it is suitable to be used in the development and utilization of the bioleaching process.Selection of refractory Shangri-La copper sulfide ore in Yunnan, after 22 days, the Cu extraction of was only 30.81%. It should be improved by using the intensified leaching method. Enhance the leaching process by adding Acidiphilium cryptum. The results showed that the reducing property of Fe3+ was weak, the Cu extraction reduce to28.14%. Add the straw cellulose treated with 50% sulfuric acid to enhance the leaching process. Adding cellulose hydrolysate can promote the leaching of chalcopyrite mineral,the Cu extraction increased by 36.35%. But the effect of the addition of cellulose hydrolysate on the leaching of pyrite is not obvious. Adding cellulose hydrolysate on leaching of copper sulfide ore from Shangri-La can increase the Cu extraction by11.91%. Exploration using stratified stacking method to deal with the low-grade ore which has a large amount of mud and small grain size, improve the permeability and efficiency of the powder ore column leaching. After 32 days stratified column leaching experiment results showed that the Cu extraction of copper sulfide ore from Shangri-La Yunnan is about 51.27%, and the Fe extraction is about 20.03%.
