Nowadays, clinic achievement of cancer therapy still fails to live up to expectations. In this paper, we respectively synthesis two types of drug delivery system, they are polyurethane based nano-carries and drug-loaded thermo-sensitive hydrogel.Drug loaded nanocarriers which based on polymeric micelles are developing in anticancer treatment. In the first part of this paper, we import piperazine group and carboxyl by polycondensation to prepare a pH-sensitive polyurethane block polymer.The experiments show that the polymer could self-assemble into micelles at pH=7.4,and the surface charge could reversal between pH 5.5-7.5. We use doxorubicin as the model drug. And the drug release profile shows that the amount of released drug increased when pH decreased. At the same time, we find that the charge reversal property could improve the endocytosis and the drug release when we experiment on HeLa cell line.The stimulus responsive materials were deep discussed in recent years, and increasingly studies are pointing in the intelligent hydrogels. Poly(amino acid) is a kind of polymer with excellent biocompatibility, and plays a unique role in drug controlled release system. The second part of this paper, we synthesis the thermo-sensitive hydrogel based on Poly(L-glutamic acid ethyl ester) to encapsulate anticancer drug using for site-specific anti-tumor therapy. The material of hydrogel is easily synthesized and molecular weights can be controlled. The intensity of the hydrogel can achieve 2000 Pa, and the transition temperature is 25 oC. The hydrogel is non-toxic. The hydrogel loaded CA4 P and CDDP simultaneously. The experiments show that the synergic effect is obviously when combined the two drugs.
