Fructose.is a new type of functional sugar source. It has pure sweet taste and high nutritional value. Fructose has been widely used in the pharmaceutical industry. In recent years, the development and utilization of fructose has drawed scientists’attention widely. The relevant research is very active. However, the research of this field develops slowly and the technology of fructose preparation can’t meet the demands of market in China.The main research of this paper included analyzing the ingredient of raw material and determined the purity of inulin. Then inulin was hydrolyzed by acid-catalyzed under microwave irradiation and its operating conditions were optimized. Finally, the chromatographic separation technology was applied to separate fructose and glucose and its operating conditions were optimized. The results of the study were as follows.1.The inulin raw materials included 98.2% total carbohydrate,7.4% reducing sugars, 0.2% ash, and 1.6% protein. So the purity of inulin was 90.8%.2.The fructose production rate under microwave irradiation was studied from concentration of inulin, temperature, pH, and reaction time. The optimal reaction condition was:inulin solution 15% , temperture 55℃, pH 5 and reaction time 15min. Under those conditions above, the rate of fructose production was 92.3%. Compared with conventional heating, micorwave irradiation not only accelerate acid hydrolysis of inulin, but also decrease the by-product and result in a higher output.3.Comparing the advantages and disadvantages of 001 x 7 type, D001 type,001 type acidic cation exchange resins, the 001 x 7 type was choosed for the separation of fructose and glucose. Through single factor test and orthogonal test, the optimal separation condition was established:feed volume 20mL, washing rate 4mL/min, column temperature 75℃, the height-diameter of the column 14. Under those conditions above, the purity of fructose was 98.5%.
