The dried root and rhizomes of four species, Gentiana manshurica Kitag., Gentiana scabra Bge., Gentiana triflora pall. and Gentiana rigescens Franch., are officially listed in the Chinese Pharmacopoeia under the name of Radix gentianae. The first three species are distributed in northeastern China and called "Guanlongdan" while the latter one is located in southwestern areas and called "Jianlongdan". It is bitter-tasted and cold-natured. It is quite frequently used as ingredients in Chinese herbal medicines for the treatment of damp-heat, jaundice, congestion, deafness, hepatitis, pains, rheumatism, and some cases for women’s diseases.The subject research systemiced about the history of processing root gentianae and its mordon literature, summed up some processing methods and aims by studying literature and compared all kinds of processing methods to establish the reasonable modern standardized processing method.Keeping to traditional processing experince of Chinese Materia Madica, the author had studied processing technics of the root and rhizomes of Gentiana scabra Bge.. Single factor experiments and orthogonal design experiments were used to optimize the processing technical parameters by evaluation of the content of the water extracts and gentiopicroside. The optimal procedure was suggested as follows: Gentiana Radix was cut into 5-10 millimetres’long, added one-fifth amounts of wine by gentian’s weight (V/W), moistened for half an hour, and then dried by slow fire at 120℃for two munites. The technics was scaled up in production and the results indicated that the processing technics were reasonable and feasible.The extracting method and chromatographic condition for the HPLC fingerprint of gentian were established, and also the typic mode of HPLC fingerprint for the drug and processed products.
