Study On Breeding Selection Of Elite Lines For Flue-cured Tobacco Based On Electron-beam Irradiation

In order to screen elite flue-cured tobacco germplasm the mutagenesis method was used on flue-cured tobacco lines Songyan A, Songyan B and Songyan C. The germination and the seedling growth condition on salt culture media were investigated. The field performance of the flue-cured tobacco seeds which are A500, B500 and C500 were also evaluated and some individual plants were selected as M1 generation seeds sources. The research results indicated that:(1) The quality of cured leaf of A is excellent among all the three flue-cured tobacco lines. The equilibrium moisture content is high, protein content is low, moderate, nicotine content and high potassium content of total sugar, carotenoid and chlorophyll content is higher. A has the best physical properties, chemical components and aroma constituents.(2) The seeds treated with 500 GY can germinate normally. More than half of the A500 seeds which is treated seeds from A germinated normally while lower than 50% of the B500 and C500.(3) The salt media which contain 500 and 1000 m M salt concentration inhibited the growth of the flue-cured tobacco seedlings. Very few of seedlings had the normal growing condition on the lower concentration of salt which is 250 m M.(4) The A500 seeds after radiation planted in fields of Guangdong have good field performance compared with the other radiation treated seeds. Results suggested that the cured leaf of Songyan A is excellent and the radiation treated seeds A500 growing well in field and with a strong resistance to disease. A500 seeds can be used to screen the excellent germ line with good quality as well as strong resistance to drought and disease.
