| Objective: The aim was to establish and evaluate the temporal lobe epilepsy(TLE) rat model which kindled by kainic acid(KA), observe the effects of alpha-asarone on the TLE model and analysis it’s mechanism. Method:SD rats were randomly divided into normal control group, epilepsy model group and treated group. The normal control group and the epileptic model group were treated with normal saline,the treated group was treated with alpha-asarone,after 15 minutes epilepsy was induced by kainite in both of the epilepsy model group and the treated group. The changes in epileptic behavior and EEG among three groups were compared, the histopathological change of hippocampus was investigated by HE staining and electron microscope, the activity of SODã€MDAã€P-AKTã€XIAPã€CASPASE-9 in rat brain was investigate. Results: 1.behavior: The number of epilepsy group rats which developed seizures at stageâ…£was 80%, the average time to developed seizures was 35.4±12.61min, the treated group showed a prolonged latent period to 65.8±10.85min and reduced the stage of epileptic behavior. 2.EEG:The normal control group could see the wave ofαandβ; the epileptic model group could see the continuous slow wave of high amplitude of wave, sharp wave, spike wave or spike slow wave discharges; the treated group’s EEG approximately normal, including the little low amplitude of sharp wave, spike slow wave discharges. 3. Pathology: The normal control group’s nerve cells are arranged tide, cell contour clear, Cytoplasmic rich, basophilic, the nucleus round, center, nucleoli clear. The epileptic model group’s neurons boundaries are not clear, cell bodies change into a circle or triangle, shrinks and smaller, cytoplasm like empty bubble, nuclear hyperchromatic, nucleoli disappear. nerve’s surrounding gap increases, could see the hole lefted by the disappear of nerves. The treated group’s number of nerve cells die and disappear was much less. Electron microscope: The normal control group’s nerve cells nuclear membrane structure is clear, chromatin is uniform, mitochondria, googi apparatus, lysosome and rough endoplasmic reticulum are completed. The epileptic model group’s neuron degeneration, shrinks, the body decreases,Lm.result fuzzy, Nuclei density, chromatin condensed and gathered to edge,most nuclei clumped chromatin and become dark,lining system expansion or change into vacuolated,mitochondria swelling or destruction, ridge disordered or break, some like empty bubble. The treated group’s nucleus structure is nearly complete, nuclear membrane structure is clear, chromatin condensed and gathered to edge a litter, lining system expansion or change into vacuolated, mitochondria swelling or destruction, ridge disordered or break, some like empty bubble. 4ã€SODã€MDA:compared with normal controls group, the model group’s MDA content is much higher,the SOD activity is much lower(P<0.01); the treated group’SODã€MDA contents are quite different from the model group (P<0.01);4.p-Aktã€caspase-9ã€XIAP: compared with normal controls group, the model group’s content of p-AKT and caspase-9 is increased (P<0.01),the content of XIAP is decreased(P<0.01); compared with the model group, the treated group’content of p-AKTã€XIAP is increased (P<0.01), the content of XIAP is decreased (P<0.01).Conclusion: 1.Seizures were induced and to establish the rat epilepsy model with unilateral intrahippocampal kainic aicd(KA) injection by stereotactic technique, the model can simulate the human temporal lobe epilepsy(TLE) feature well and is the ideal TLE model. 2.Alpha-asarone can prevent epileptic seizures in some extent, improved seizure outcome, is a good drugs to cure temporal lobe epilepsy. 3.The alpha-asarone can remove excessive free radicals produced in the process of seizures, reduce the lack of oxygen in some place, prevent excessive damage of brain cells, it can also affected the PI3K/AKT signaling pathways, regulation the expression of XIAPã€caspase-9 in mitochondrial way, play neural protection role by antiapoptotic,promote live,so that the alpha-asarone can inhibit abnormal discharge, treatmented epilepsy. |
