Studied On The Synthesis Of Voriconazole

This thesis aims at the studies on the total synthesis of voriconazole, which consists of the following two chapters:Chapter 1. total synthesis of voriconazoleTwo routes toward total synthesis of voriconazole were designed and tried. Finally, the total synthesis of voriconazole has been achieved in seven steps, which involved hydrogenation、bromation、friedel-crafts acylations、Refonnatsky reaction、epoxidation、ring-opening reaction under the alkaline condition, resolution of compound 11 with asβ-NSA we can get the voriconazole. But as the unstabitily of 11 under this condition, the yield was low. The relative work is still going on.Chapter 2. review about Reformatsky reactionThis review has covered the history of the asymmetric Reformatsky development. The preparation, mechanism of Reformatsky and application in natural products total synthesis of Reformatsky were described in detail.
