Objective①We detect the expression of Anti-mullerian hormone (AMH) in patientswith polycystic ovary syndrome (PCOS),premature ovarian failure (POF) and normal controlsin order to clarify the relationship between serum AMH levels and ovarian reserve;②Thisstudy investigate the AMH and AMH type receptorII(AMHRII)genotype distribution inpatients with PCOS、POF patients and normal control group aimed to elucidate the molecularmechanisms in genetic level.Methods①Serum levels of AMH in32cases of PCOS,25cases of POF,30cases of normalcontrol group were determined by using enzyme-linked immunosorbent assay (ELISA), at thesame time we measured serum follicle stimulating hormone(FSH),luteinizinghormone(LH),estradiol(E2),testosterone(T) and prolactin(PRL) levels by using biochemicalmethods,and recorded the indexs,such as participant age,body mass index(BMI),fastingglucose and insulin levels,2-hour postprandial insulin levels;②Polymorphisms and genotypeof AMH and AMHRⅡ were determined by polymerase chain reaction(PCR) and DNAsequencing in94cases of PCOS patients,32cases of POF patients and94cases of normalwomen,while compare age,BMI and hormone levels in different distribution of AMH andAMHRⅡgenotypes.All statistics are using SPSS13.0measurement data with the t-test,countdata with the2test,the correlation between various indicators by Pearson analysis.Results①The expression of AMH in serum is significantly increased in PCOS group,however,the expression of AMH is obvious lower in POF group compared with control group (P<0.05);b.The expression of serum FSH,LH,PRL,E2,T and AMH levels were statisticallysignificant among the three groups(P<0.05),but the indexs of age and BMI have no significantdifference(P>0.05)in the three groups c.Only serum AMH and FSH in the POF shows anegative correlation (P<005);d.In the PCOS The expression of AMH in serum was positivelycorrelated with L,T,BMI,postprandial2hours of insulin level(P<0.05),but was not correlatedwith FSH and insulin resistance index;②a.The distribution of AMH,AMHRⅡ genotype andAMH allele frequency were significant difference(P<0.05) in three groups,but the AMHRⅡallele frequency was not significant difference(P>0.05);b.In different distribution of AMHgenotype BMI,LH,T were significant difference in PCOS(P<0.05),while it was not significantdifference in the normal group and POF(P>0.05);c.In different distribution of AMHRⅡgenotype BMI,T,E2were significant difference in POF(P<0.05),while age,BMI,FSH,LH,T,E2were not significant difference in PCOS and normal group(P>0.05).Conclusions①Serum AMH expression is difference in the different ovarian reservefunction,it may be involved in PCOS occur,development,and sex hormone synthesis disordersrelated;②a.Ovarian reserve function may be relevant with AMH and AMHR Ⅱ genepolymorphism;b.The levels of BMI,LH,T in PCOS patients may be relevant with the AMHgene polymorphism;c.The level of BMI,T,E2in POF patients may be associated with AMHRⅡ gene polymorphism.
