| OBJECT: Renal interstitial fibrosis (RIF) is a common way that variety of chronic kidney diseasessprogress to the end-stage. Therefore, the prevention and cure of RIF has a great signification tothe chronic kidney disease. The Renal Tubular Epithelial Cells has played an important role in thefibrosis progress, The pathological changes of Tubulointerstitial are the main factors ofdetermining the chronic renal disfunction process, which have a complex trigger and developingfundamental. The Jiedu Fushen Zhuyu Detoxification is an efficacious detoxified description thatsums-up by the long-term clinic practice by Fujian Provincial Hospital. The main purpose of thisstudy is to discuss the effect and the possible mechanism of this description, provide the labevidence to the application of the description, and offer the new idea and proofs for the RIFtreatmentMETHOD: we established the transforming growth factor β1(TGF-β1) induced fibrosis model ofhuman Renal Tubular Epithelial Cells,Then Using MTT, WesternBlot, invasion assay and othermethods observed the influence of Jiedu Fushen Zhuyu Detoxification on the cell proliferation,the changes of extracellular matrix and the expression of connective tissue growth factor. Afterthat, by the use of rat model of unilateral ureteral ligation (UUO), to observed the medical effect invivo and verified the possible effect concluded by the cellular experiment.RESULTS: The cellular experimental results show that:After cultured3d with HK-2cells, there’s visible cell morphological changes, that becomehypertrophic, elongated into a spindle-like myofibroblasts, and a numbers of HK-2cells expressingα-SMA, collagen type Ⅲsecretion increase, CTGF expression was significantly increased as well.The Jiedu Fushen Zhuyu Detoxification could restrain the morphological changes in HK-2cellsinduced by TGF-β1. The majority cells maintain the normal epithelial cells statues. At the sametime, the α-SMA positive cells decreased, and showed a dose-dependent association. Each groupof the drug intervention Ⅲ collagen content were lower than TGF-β-induced group, and showed adose-dependent association. Meanwhile, the intervention group of drugs can significantly reduceTGF-β1-induced CTGF expression, explaining that inhibition of CTGF expression could be the keytarget of blocking TGF-β1-induced transdifferentiation.Animal experiments show that the Jiedu Fushen Zhuyu Detoxification could significantly reducedthe level of renal interstitial fibrosis in the UUO model in rats. Histopathological andimmunohistochemical analysis showed that the retaining of the renal fibrosis was related to theregulation of MMP-2, and the balance between MMP-9and TIMP and the inhibition of CTGFexpression.The results is consistent with the cells experimental.CONCLUSION: The Jiedu Fushen Zhuyu Detoxification could reduce the damage of the renalinterstitial fibrosis, and may relate to the regulation of TGF-β1-CTGF signal pathway. The Animalexperiments further approved the effect of the drugs, and indicated that the effect of the drugs wasrelated to the balance of MMP/TIMP. |
