| Objective To investigate the effect of nesfatin-1on gastric motility via recording the regulation of nesfatin-1on the contractivity of gastric smooth muscle strips and gastric emptying in rats.Methods By isometric force transducers connecting with electrophysiolograph, the regulation of0.026μmol/Lã€0.26μmol/Lã€2.6μmol/L nesfatin-1on the contractivity of gastric fundus〠gastric corpus and gastric antrum circular smooth muscle strips in male Wistar rats was measured. The control group were given the same dose of physiological saline. Insert a tube into DVC and inject respectively0.5μmol/Lã€5.0μmol/Lã€50.0μmol/L nesfatin-1into it, observe its impact on gastric emptying. The control group were given the same dose of physiological saline.Results In the gastric smooth muscle experiment,(1) the tensile force of gastric fundus smooth muscle strips in the0.026μmol/L,0.26μmol/L,2.6μmol/L nesfatin-1was obviously decreased(n=10,P<0.01). Tension percentage changes are (84.78±7.90)%,(76.20±8.50)%and (62.65±6.90)%. The effect was dose-dependent (P<0.05~0.01);(2) the tensile force of gastric corpus smooth muscle strips in the0.026μmol/L,0.26μmol/L,2.6μmol/L nesfatin-1was obviously decreased(n=10,P<0.01).Tension percentage changes are (84.87±8.10)%,(75.96±7.70)%and (63.78±7.90)%. The effect was dose-dependent(P<0.05-0.01);(3) the tensile force of gastric antrum smooth muscle strips in the0.026μmol/L,0.26μmol/L nesfatin-1was not significantly decreased (n=10,P>0.05). Tension percentage changes are (92.59±13.06)%and (90.76±10.11)%. The tensile force of gastric antrum smooth muscle strips in the2.6μmol/L nesfatin-1was obviously decreased(n=10,P<0.01). Tension percentage change is (77.21±13.48)%. Respectively injecting0.5μmol/L,5.0μmol/L,50.0μmol/L nesfatin-1into DVC induced a reduction of gastric emptying(n=8,P<0.05~0.01). The effect was dose-dependent(P<0.05~0.01).Conclusion Nesfatin-1inhibits the contractivity of gastric smooth muscle strips and steps down gastric emptying. |
