The Study Of Correlation Between Pancreatic Carcinoma And Single Nucleotide Polymorphism Of β2-adrenergic Receptor Gene

Objective:To investigate the correlation between the clinicopathologic characters, the expression of β2-AR, VEGF, MMP-2protein and the single nucleotide polymorphism (SNPs) in position16and27of β2-adrenergic receptor (β2-AR) gene in pancreatic carcinoma (PC), and explore the clinical significance.Methods:(1) DNA of pancreatic carcinoma group (PCG) and non-neoplastic control group (NCG) were extracted from FFPE tissue sections, using a Qiagen QIAamp DNA FFPE Tissue Kit.(2) Genotype of gene (32-AR were conducted by PCR-restriction fragment length polymorphism (PCR-RFLP) and DNA sequencing.(3) The expression of (32-AR, VEGF, MMP-2protein was detected by immunohistochemistry in tissue specimens of pancreatic carcinoma.(4) The X2test was performed for nonparametric comparisons of data to investigate the correlation between the clinicopathologic characters, the expression of protein and the SNPs in β2-AR gene in pancreatic carcinoma using software from the spss17.0statistical program package.Results:(1) The difference in distribution of the genotypes and alleles between PCG and NCG is insignificant (P>0.05).(2) The genotype frequencies of position16had no significant correlation with the gender, age, histological differentiation and size of tumor (P>0.05),they had significant correlation with lymph node metastasis,TNM grade, one year of survival and the expression of β2-AR, VEGF, MMP-2protein (P<0.05).(3) The genotype frequencies of position27had no significant correlation with clinicopathologic characters, and the expression of β2-AR, VEGF, MMP-2protein (P>0.05).Conclusion:It may be helpful to assess lymph node metastasis and prognosis of pancreatic carcinoma by detecting SNPs in position Argl6Gly of P2-AR gene. G allele of position16indicates high risk of lymph node metastasis, high TNM grade, low one-year survival and elevated protein expressions of β2-AR, VEGF and MMP-2.
