Background Lung cancer have high incidence and mortality.Although we have many therapeutic methods,the effectiveness and the quality of life is unsatisfactory.The theory of cacer stem cell brings many experiment and clinical study.Scientists have constinuously sorted cancer stem cells for many kinds of tumors according to the protein marker on cell surface.However,nonspecific lung cancer stem marker so far.At present,many researches reports that CD133(+)CD34(+)have the ability of tumor formation in vitro and in vivio,but combined with ALDH1A1is not commen.Objective To study the expression of cancer stem marker CD133,CD34and ALDH1A1in lung tumor and search the relationship of the markers expression between clinical festures.Methods A total of68cases lung tumor under operation in Yuhuangding Hospital from Jan.2010to Apr.2011with complete clinical follow-up data was involved in this study.In which,50are lung caner and18are benign tumor of lung.To study the expression of CD133,CD34and ALDH1A1by using immunohistochemistry, we statistically analysed the expression of CD133,CD34and ALDH1A1to search the relationship of tumor marker and patient-related information. Results1.CD133The exepression rate of CD133in lung cancer cases was54%(27/50),which were all significantly hinger than those in benign lesions10%(2/18) and in tumor-adjacent normal tissuess6%(3/50). CD133expression was related to the diffrention degree of lung cancer.CD133positive expression was associated with poor prognosis of lung cancer patients. The positive expression rate of CD133in the patients with low-grade differentiation was higher than that with high-grade and middle-grade differentiation, they were71.4%(15/21)and41.4%(12/29) respectively (X2=4.428,P=0.035).There was no significant differences between the expression of CD133and the age, smoking index, tumor size, TNM stage, lymphoid metastasis as well as histological type.2. CD34The MVD in lung cacer cases was20.16±6.398,which were all signifi-cantly hinger than those in benign lesions (14.40±3.050) and in tumor-adjacent normal tissuess (11.76±3.642). CD34expression was related to tumor size, TNM stage, differentiation and lymphoid metastasis of lung cancer.MVD was associated with poor prognosis of lung cancer patients. The MVD in tumor size with>3cm was higher than that≤3cm, they were23.00±5.24and19.35±5.75respectively (X2=-2.342, p=0.023). The MVD in the patients with I and II stage was lower than that III and IV grade, they were19.00±5.34and25.18±4.22respectively (X2=-4.144P=0.000). MVD in the patients with low-grade differentiation was higher than that with high-grade and middle-grade differentiation, they were23.29±3.94and19.51±6.38respectively (X2=2.392,P=0.021). The MVD in the patients with lymphoid metastasis was higher than that no lymphoid metastasis, they were23.72±4.61and18.48±5.67respectively (X2=3.586,p=0.001).There was no significant differences between the expression of CD34and the age, smoking index, as well as histological type.3. ALDH1A1The exepression rate of ALDH1A1in lung caner cases was40%(20/50),which were all significantly hinger than those in benign lesions(0) and in tumor-adjacent normal tissuess(36%,9/25). ALDH1A1expression was related to the TNM stage of lung cancer.ALDH1A1positive expression was associated with poor prognosis of lung cancer patients. The positive expression rate of ALDH1A1in the patients with I and II stage was higher than that III and IV stage, they were64.7%(11/17) and27.3%(9/33) respectively (X2=6.551, P=0.010).There was no significant differences between the expression of ALDHlAl and the age, smoking index, tumor size, lymphoid metastasis, differentiation as well as histological type.Conclusion1. The expression of CD133is related with tissue differentiation-state.2. CD34expression was related to tumor size, TNM stage, differentiation degree differentiation and lymphoid metastasis of NSCLC.3. ALDH1A1expression was related to the TNM stage of NSCLC.4.CD133,CD34and ALDH1A1can be seen as postoperative risk factrors.5. CD133,CD34and ALDH1A1can be used as appropriate combination of surface markers of cancer stem cells for human lung carcinoma.
