| Objective:Hydronephrosis is a kind of clinical symptom of internal and external factors acton urinary tract,if the factors causing hydronephrosis can not be released,the kidneywould be ischemia and hypoxia because of the oppression of urine.it is significance torelease the obstacle as soon as possible for protect the renal.The study found theoccurrence of vascular primarily through the VEGF signaling pathway.Recently thestudy have found that miRNA can regulate angiogenesis, HIF would be upregulationand some of miRNA would be expressed significantly after ischemia or hypoxia.Research also suggests that miRNA-210may influence angiogenic by regulatingdownstream genes.This study is mainly observe the expression of inducible factor-1(HIF-1)vascular endothelial growth factor(VEGF) microRNA-210and explore the regulatorymechanism after unilateral ureteral occlusion (UUO).Methods:the UUO models are made by ligation unilateral ureteral with silk,and theexperiment divided into sham and uuo,All the mice were sacrificed by cervicaldislocation after2,5or14days of UUO.the kidney samples were examined for HIF-1mRNA,VEGF mRNA and miR-210by quantitative Real-Time reversetranscription-polymerase chain reaction(RT-PCR),HIF-1protein by Western-blotting.Results:the expression level of HIF-1mRNA was gradually raised after UUO,Comparedto the sham group.the expression level of VEGFmRNA and microRNA-210is upregulation after2days of UUO and down regulation after5,9or14days of UUO,Compared to the sham group.the expression level of HIF-1protein increased afterUUO Compared to the sham group.Conclusion:the results of the level of HIF-1protein increased and VEGFmRNA and microRNA-210up regulation temporary may related to the adaptive response ofkidney hypoxia and ischemia after UUO. |
