| Objective:By detecting the expression of Nrf2and HO-1in the colon cancer tissues and thenormal colon tissues adjacent to carcinoma, explore the colon cancer with bleedingwhether impacting on the expression of these two proteins, and analyze therelationship between their expressions and clinicopathological parameters, in order tofind the specific diagnostic marker and effective therapeutic targets of colon cancer.Methods:Detection of Nrf2and HO-1expressions in57cases of colon cancer tissues and20cases of corresponding normal colon tissues adjacent to carcinoma (from foci edge>5cm) by immunohistochemistry, observe the differences of these two proteinsexpression between the colon cancer bleeding and non-bleeding environment, andanalyze the relationship between their expressions and main clinicopathologicalparameters. The test results are used SPSS19.0statistical package for statisticalanalysis.Results:1. The expression of Nrf2and HO-1in colon cancer: the positive expression rateof Nrf2in colon cancer was73.68%(42/57)which was significantly higher than thatof30.00%(6/20) in normal tissue adjacent to carcinoma, the difference had statisticalsignificance (P<0.01); the positive expression rate of HO-1in colon cancer was70.18%(40/57)which was significantly higher than that of25.00%(5/20)in normaltissue adjacent to carcinoma, the difference had statistical significance (P<0.01).2. The influence of colon cancer with bleeding to the expression of Nrf2andHO-1: the positive expression rate of Nrf2was89.29%(25/28)in colon cancerbleeding group, and the positive expression rate was58.62%(17/29)in colon cancernon-bleeding group, their difference had statistical significance (P<0.01); the positiveexpression rate of HO-1was85.71%(24/28)in colon cancer bleeding group, and thepositive expression rate was55.17%(16/29)in colon cancer non-bleeding group, theirdifference had statistical significance (P<0.05). 3. Association of Nrf2and HO-1expressions with main clinicopathologicalparameters: the positive expression rate of Nrf2was66.67%(14/21) in preoperativeCEA<6.5ng/ml group, and the positive expression rate was77.78%(28/36) inpreoperative CEA≥6.5ng/ml group, the difference had no statistical significance(P>0.05); the positive expression rate of Nrf2was60.61%(20/33) inwell-differentiated group, and the positive expression rate was91.67%(22/24)inpoorly differentiated group, the difference had statistical significance(P<0.01); thepositive expression rate of Nrf2was92.59%(25/27)in tumor metastasis group, andthe positive expression rate was56.67%(17/30)in non-tumor metastasis group, thedifference had statistical significance (P<0.01); In Dukes stage, the positiveexpression rate of Nrf2was53.33%(16/30) in A+B stage, and the positive expressionrate was96.30%(26/27) in C+D stage, the difference had statistical significance(P<0.01). The positive expression rate of HO-1was61.90%(13/21)in preoperativeCEA<6.5ng/ml group, and the positive expression rate was75.00%(27/36) inpreoperative CEA≥6.5ng/ml group, the difference had no statistical significance(P>0.05); the positive expression rate of HO-1was54.55%(18/33) inwell-differentiated group, and the positive expression rate was91.67%(22/24)inpoorly differentiated group, the difference had statistical significance(P<0.01); thepositive expression rate of HO-1was85.19%(23/27)in tumor metastasis group, andthe positive expression rate was56.67%(17/30)in non-tumor metastasis group, thedifference had statistical significance (P<0.05); In Dukes stage, the positiveexpression rate of HO-1was50.00%(15/30) in A+B stage, and the positiveexpression rate was92.60%(25/27)in C+D stage, the difference had statisticalsignificance (P<0.01).4. In57cases of colon cancer tissues, there were38cases of Nrf2and HO-1Positive expression and13cases of Nrf2and HO-1negative expression at the sametime, there were4cases of Nrf2positive expression but HO-1negative expression,there were2cases of Nrf2negative expression but HO-1positive expression. Afterstatistics analysis, the result showed that Nrf2and HO-1expression in colon cancerhad a significantly positive correlation (rs=0.743, P<0.01). Conclusion:1. Nrf2and HO-1were highly positive expression in colon cancer tissues, theywere significantly higher than in adjacent normal colon tissues, it suggested that theexpression of these two proteins were associated with the development of coloncancer.2. The expression of Nrf2and HO-1was significantly increased in colon cancerwith bleeding, it suggested that the bleeding may through cause iron overload, induceoxidative stress response and prompt Nrf2and HO-1expressions to affect thedevelopment of colon cancer.3. The expression of Nrf2and HO-1had no correlation with the level ofpreoperative CEA high or low, but there was a correlation with pathological grade,lymph node metastasis and Dukes stage of colon cancer.4. Along with colon cancer pathologic stage, lymph node metastasis and Dukesstage degree increasing, the positive expression rate of Nrf2and HO-1alsoaccordingly increased, it suggested that the expression of Nrf2and HO-1hadcorrelation with progression and metastasis of colon cancer, there had an impact onthe prognosis of patients.5. The expression of Nrf2and HO-1had correlation,it suggested that they play animportant role in oxidative stress which caused by colon cancer,and there may have asynergistic effect to promote the development of colon cancer. |
