Objective:Detection of homeobox genes D8(HOXD8) differentially expressed inhuman ovarian cancer SKOV3and SKOV3-DDP cell line to explore the relationshipbetween HOXD8cisplatin resistance genes and ovarian cancer.Methods: Cultured human ovarian cancer cell line SKOV3and its pairedcisplatin-resistant cell lines SKOV3-DDP, application XTT colorimetric assay SKOV3cell lines and cell lines SKOV3-DDP cisplatin IC50, and calculate the resistance index;semiquantitative RT-PCR and quantitative PCR to detect differences HOXD8mRNAexpression in cell lines SKOV3and SKOV3-DDP cell lines; applications Elisadifferentially expressed proteins were detected HOXD8cell lines SKOV3andSKOV3-DDP cell lines.Results:1.Cells were treated with different concentrations of cisplatin72hours aftertreatment with increasing concentrations of cisplatin, inhibition rate increase, asignificant dose-dependent, and resistant cell line by cisplatin, the inhibition rate wassignificantly lower than the sensitive strain cell lines (P <0.05). XTT colorimetric resultsshowed, SKOV3cell lines to cisplatin IC50value of2.462±0.81ug/ml, SKOV3-DDPcell lines to cisplatin IC50value of9.174±0.24ug/ml, index SKOV3-DDP-resistant celllines was3.73, can be resistant to experiment.2. Semi-quantitative RT-PCR results showed, SKOV3cell lines HOXD8mRNAexpression level was0.238±0.037, SKOV3-DDP cell lines HOXD8mRNA expressionlevel was0.707±0.026. SKOV3cell lines compared with the relative expression celllines SKOV3-DDP amount HOXD8mRNA was significantly increased, the differencewas statistically significant (P <0.05). Quantitative PCR results showed, SKOV3celllines HOXD8mRNA expression level of0.1711±0.026, SKOV3-DDP cell linesHOXD8mRNA expression level was0.5761±0.037. SKOV3cell lines compared with the relative expression cell lines SKOV3-DDP amount HOXD8mRNA was significantlyincreased, the difference was statistically significant (P <0.05).3. Elisa results show, SKOV3cell lines for protein expression levels HOXD81584.4±2.08pg/ml, SKOV3-DDP cell lines HOXD8mRNA expression level was4423.3±1.52pg/ml. Compared with SKOV3cell lines, SKOV3-DDP cell lines expressing theprotein significantly increased the amount of HOXD8differences were statisticallysignificantly (P <0.05).Conclusions:1. HOXD8mRNA and protein in cultured HOXD8ovarian cancer cell lineSKOV3-DDP and SKOV3cell lines were expressed.2. SKOV3-DDP cell lines HOXD8expression was significantly higher than that inSKOV3cell lines, and its expression increases there is a clear correlation withcisplatin-resistant ovarian cancer.
