| Objective: To investigate the role of IL-1β in capillary leak syndrome by observingthe AQP-1expression, apoptosis and ultrastructural of vascular endothelial afterIL-1β intervention.Methods: Umbilical vein endothelial cells (UVEC)in vitro were randomly dividedinto3groups including Time groups, in which UVEC were treated with20ng/mlIL-1β for3h(T1),8h(T2),12h(T3), or24h(T4) respectively, Concentration groups, inwhich UVEC were treated with0.2ng/ml(C1),2ng/ml(C2) or20ng/ml(C3) IL-1βrespectively for24h, and Control groups, in which UVEC were incubated with culturemedium without IL-1β for24h. The changes of AQP-1mRNA and protein expressionwere detected by real-time PCR and western blot. Apoptosis was analized by flowcytometry. Cell ultrastructural changes were observed by electron microscopy.Results: AQP-1mRNA and protein expression of T1~T4groups and C1~C3groupswere lower than that in Control group (P <0.05). Apoptosis rate was increased, andmitochondrial swelling, vacuolar degeneration, karyolysis and necrosis were observedunder electron microscopy. All the changes above could be observed in a dose-andtime-dependent manner.Conclusions: IL-1β can decrease AQP-1mRNA and protein expression, increaseapoptosis rate and damage cells ultrastructure in UVEC. It’s important for vascularendothelial barrier damaged and may be associated with capillary leak syndrome. |
