Objective:To study the relationship between the Golph3expression and theclinicopathological parameters, cell proliferation, apoptosis orprognosis in colorectal canecr(CRC) tissues.Methods:The expression of Golph3was examined by western blotting in62casesof colorectal cancer tissues and colorectal mucosae, immunohistochemicalstaining of mTOR, Ki-67,Golph3and Caspase-3was performed by the standardstreptavidin-peroxidase (SP) technique for the paraffin sections of62colorectal cancers. Apoptotic index in CRC was examined by TUNEL technique.The relationship between the Golph3expression and the clinicopathologicalparameters, the mTOR expression,proliferation index and apoptosis indexwas analyzed. The correlation between Golph3expression and the prognosisof patient was analyzed with the Kaplan-Meier statistical method.Results:1. The expression of Golph3in colorectal carcinoma tissuesBy western blotting test, compared with normal colorectal mucosae(0.61±0.53), the notably higher level of Golph3expression was confirmed incolorectal cancer tissues at protein level(0.76±0.59)(P<0.05). With thestandard streptavidin-peroxidase (SP) technique, staining of Golph3was located in the cytoplasm, the positive rate of Golph3expression inthe CRC tissue (53.2%) was significantly higher than in the normalcolorectal mucosae (24.2%)(P<0.05). Likewise, the Golph3expression in poorly differentiation, outside serous membrane, lymph node metastasis andstage Ⅲ of CRC was significantly higher than in middle to welldifferentiated, inside serous membrane, no lymph node metastasis and stageⅠ-Ⅱ of CRC(P<0.05). However, there was no correlation between Golph3expression and other clinicopathological parameters such as age, sex,tumor site, tumor depth and length(P>0.05).2. Correlation between Golph3expression and mTOR expression,proliferation index in colorectal cancer tissuesThe protein level of Golph3expression in mTOR-positive colorectalcancer(0.95±0.73) was significantly higher than in mTOR-negativecolorectal cancer(0.54±0.28)(P<0.05), and the protein level of Golph3expression(0.76±0.59) positively correlated with Ki-67proliferationindex(0.46±0.17) in colorectal cancer via Pearson correlation analysis(r=0.325,P=0.010).3. Correlation between Golph3expression and caspase-3expression,apoptotic index in colorectal cancer tissuesThe caspase-3expression and apoptotic index in Golph3-positive CRCtissue were significantly lower than in Golph3-negative CRC(P<0.05), andthe Golph3expression negatively correlated with apoptotic index in CRCvia Spearman correlation analysis(r=-0.320,P<0.05).4.The relationship between the Golph3expression and prognosis.The5-year survival rate in Golph3-negative colorectal cancer (69.4%)was significantly higher than that in Golph3-positive colorectalcancer(48.6%)(Log-rank test,P<0.05).Conclusions:A high expression of Golph3was found in colorectal carcinoma tissues,and it was correlated with the degree of differentiation,invasion depth,lymph node metastasis and TNM staging. Overexpression of Golph3may promotecell proliferation and inhibite cell apoptosis. Otherwise, our data suggest that Golph3expression may be a novel prognostic marker for patientwith colorectal cancer.
