| With the improvement of people’s living standards, under the influence of poor eatinghabits and the change of dietary structure, obesity and hyperlipidemia patients is increasing, anddirectly lead to a high risk for the sharp rise in the incidence of high blood pressure,arteriosclerosis andâ…¡ diabetes which could harm to people’s health.Therefore, on the basis of notrestricted diet and non-toxic side effects, to prevent blood lipid metabolic abnormalities,hyperlipidemia and obesity in efficient and safe ways are pay more attention to highly. Velvetantler commonly used for nourishing health care medicine in our country has a history of twothousand years, was recorded in many prescription and belongs to the non-toxic drug, and it willdoes not cause side effects as long as in the rational uses.At present, there is a lot of research about the extraction of velvet antler chemicalcomposition, biological activity, the use of clinical medicine and pharmacology in domestic, butthe application in lipid-lowering has reported less. Therefore looking for the compositions andmechanism of velvet antler on weight-reducing and lipid-lowering has scientific significance ofin-depth study, and also has practical significance to the prevention and treatment of obesity andhyperlipidemia.This test extracted protein and polysaccharide from velvet antler and designed by responsesurface, optimized the three influence factors in the process of extraction of velvet antler proteinand four impact factors in the process of extraction of velvet antler polysaccharide, to improvethe yield. Put the extracted protein and polysaccharide of velvet antler from this test into theanimal experiment, and set up three different doses to study the effect of lipid-lowing andweight-reducing and the mechanism of it. The results are as follows:1. This test combined the method of ethanol extraction with ultrasonic extraction, andoptimized the liquid ratio, ethanol extraction and ultrasonic extraction time and so on threefactors.The sequence of single factor’s influence on velvet antler protein yield is: liquid ratio>ethanol extraction time> ultrasonic extraction time. The best extraction conditions of velvetantler protein designed by response surface are: liquid ratio was18:1, ethanol concentration was65%, ultrasonic extracting time was21min. Under the best conditions the antler protein yield of3.2113%.2. By the combination of the methods of ultrasonic assisted extraction, hot water extractionand alkali extraction. Boxï¼Behnken experiment was also designed to set four influence factors -liquid ratio, hot water extraction time, ultrasonic assisted extraction time and alkali extractiontime and optimized the four factors. The sequence of the four single factor’s influence on velvetantler polysaccharide yield was: hot water leaching time>liquid ratio> alkali extraction time>ultrasonic assisted extraction time. Regression analysis of response surface by using thestatistical software Design-Expert, the best extraction conditions of velvet antlerpolysaccharide for the liquid was42:1, ultrasonic assisted extraction time was21min, hotwater extraction time was2h, alkali leaching time was2.3h, the antler polysaccharide yieldunder this conditions of1.0539%.3. In weight-reducing trials of obese mice, velvet antler polysaccharides obviouslydecreased obese mice’s body weight, reduce the index of fat, reduce the content of TC, TG,LDL-C in serum, raise the content of HDL-C, make the fat cells change smaller, so as toachieve the aim of weight-reducing and lipid-lowing. The mechanism may be related todecrease the content of FAS, and has nothing to do with reduced the activity of LPL. Velvetantler protein, by contrast, has no effect on lipid-lowering and weight-reducing, but the velvetantler protein and polysaccharide has increased the content of SOD, decreased the lipidperoxidation of polyunsaturated fatty acids, and lowered the content of MDA. |
