Expression And Clinical Significance Of Livin And Caspase-7in Human Osteosarcoma

Purpose:Osteosarcoma is one of the orthopedic clinical common malignant tumor, it canhappen at different ages, but mainly in children and adolescents, good hair at longbone epiphysis end, high malignant degree, easy early metastasis and poor prognosis,the treatment is currently the orthopedic community’s problems. At present, thetreatment of osteosarcoma is still operative treatment is given priority to,supplemented by radiation and chemotherapy. But in recent years with thechemotherapy drug resistance and side effects of larger, traditional treatment receivedserious challenges, make more and more biological target therapy for cancer research.The body’s cells apoptosis disorders are closely associated with the occurrence anddevelopment of osteosarcoma, is one of an important mechanism in the process offormation of osteosarcoma. How accurate osteosarcoma cells apoptosis, thoroughlyremove tumor cells, the key is to find the biological treatment of osteosarcoma genetargets. Livin protein found in recent years is one of the new members of apoptosisinhibiting protein family, its high expression in a variety of malignant tumor andtumor cell line, and low or no expression in normal tissue, can inhibit the apoptosis oftumor cells. In family of aspartic acid protease (Caspase) is a kind of in the spacestructure and the amino acid sequence has the similarity of protease. Cell apoptosis isa complex and has a procedural process, main is mediated by the Caspase familyprotease cascade reaction process, the effect of its downstream area Caspase-7is theimportant molecular cascade, is one of the major members of the body to performapoptosis. This experiment was to study Livin and Caspase-7in the positiveexpression rate of osteosarcoma tissue, and the Livin Caspase-7with osteosarcomarelationship between various clinical indicators; Preliminary analysis and thecorrelation of both positive expression in osteosarcoma tissue, further explore Livinand Caspase-7in the early clinical diagnosis, treatment and prognosis ofosteosarcoma in meaning. Methods:Collect the second affiliated hospital of nanchang university bone surgery inJanuary2001to December2012admitted during the period of paraffin specimens of51patients with osteosarcoma and12cases normal bone tissue specimens, paraffinspecimens confirmed by pathology of the first the second affiliated hospital ofnanchang university for osteosarcoma. Using immunohistochemical method testspecimens of osteosarcoma Livin and the expression of Caspase-7. ApplicationSPSS19.0statistical software for statistical analysis of the results. Positive expressionrate of two indicators chi-square test and Spearman rank correlation test, with P <0.05for the difference was statistically significant. Analysis of two indicators ofexpression and the clinical pathology, the relationship between the targetosteosarcoma and osteosarcoma tissue expression in the presence of correlationbetween the two.Results:1、livin in osteosarcoma tissue and normal expression of callus tissueLivin in51patients with osteosarcoma of the positive expression rate of66.7﹪(34/51), and in12cases of normal tissue specimens in the positive expression ratewas8.3%(1/12), the positive expression rate of difference between two groups havestatistical significance (P <0.05). Livin in the tissue of osteosarcoma positiveexpression and the tumor size, osteosarcoma Enncking stages and5years of survivalrate (P <0.05), and patients’ age, gender, pathological classification, and has nothingto do the site of tumor (P>0.05);(see table1)2、Caspase-7in osteosarcoma tissue and normal expression of callus tissueCaspase-7in51patients with osteosarcoma were positive expression rate was49.0%(25/51), and in12cases of normal tissue specimens in the positive expressionrate was100.0%(12/12), the positive expression rate of difference between twogroups have statistical significance (P <0.05).
