Objective: To study the expression of TGF-β1ã€IL-6ã€TNF-α of rat hepatic stellatecells (HSC-T6) in fluid environment containing Echinococcus multilocularis.Methods: Co-culturing rat HSC-T6cells with0,0.01,0.025,0.05,0.1,0.2,0.4,0.9,1.7,3.4,6.8,13.5mg/ml of Echinococcus multilocularis fluid respectively24h, andthen detecting the expression of transforming growth factor β1(TGF-β1), interleukin6(IL-6) and tumor necrosis factor-α (TNF-α) in each groups by ELISA method.Results: Expression of TGF-β1was slightly higher than that in the blank controlgroup when Echinococcus multilocularis fluid concentration in0.01~6.8mg/mlrange, although the difference was not statistically significant; When theconcentration of Echinococcus multilocularis fluid exceeded3.4mg/ml, theexpression of IL-6increased significantly (P<0.01),6.8and13.5mg/mlEchinococcus multilocularis fluid treatment groups compared with the blank controlgroup were increased by3028.65,3879.23folds respectively; When Echinococcusmultilocularis fluid concentration lower than0.2mg/ml, the expression of TNF-αtended to be stable, and there was a expression peak in0.4mg/ml Echinococcusmultilocularis fluid but levels have dropped since then.Conclusion: TGF-β1of rats HSC-T6cells without obvious stimulation secretion byEchinococcus multilocularis fluid treatment; Expression of IL-6may be caused by ahigh concentration of Echinococcus multilocularis fluid stimulation; And thesecretion of TNF-α may be caused by a certain concentration of Echinococcusmultilocularis fluid treatment. |
