OBJECTIVE To investigate if there are mesenchymal stem cells from human gastric cancer (hGC-MSCs) and human gastric cancer adjacent non-cancerous tissues (hGCN-MSCs). Identified hGC-MSCs and researched their biological characters such as morphological, proliferation characteristics, surface markers, multi directional differentiation potential, so as to providing research basis for the impact of hGC-MSCs on gastric cancer cells research, and providing theoretical basis for the mesenchymal stem cell clinical safety issues. METHODS Collected16cases of postoperative specimens from gastric cancer patients. Used cell adherent methods and enzyme digestion methods to isolate and culture mesenchymal stem cells from fresh gastric carcinoma and carcinoma adjacent tissue. hGC-MSCs morphology and proliferation characteristics were observed; and their growth curve were measured, analyzed and compared the growth curve between difference generation; karyotype analysis of hGC-MSCs, observed the number of chromosomes and chromosome G-banding; used flow cytometry to detect hGC-MSCs surface markers expression; and induced hGC-MSCs differentiated into lipocyte cells and osteoblast cells to identify the hGC-MSCs’potentiality of multi-directional differentiation. RESULTS hGC-MSCs can be successfully isolated and subcultured by two methods in vitro, but failed to isolated hGCN-MSCs from gastric cancer adjacent non-cancerous tissues. Cells were spindle-shape and polygon as the fiber-like cell under inverted microscope. While cells reached confluence, cells arranged regularly, showed whirlpool-shape or radial-shape. Cell growth curve showed that the proliferation ability of hGC-MSCs is quite strong and declined with passages, the passage3hGC-MSCs’growth curve by the two isolation methods obtained were no significant difference, but passage18hGC-MSCs’proliferation ability has been reduced compared passage3hGC-MSCs; Karyotype analysis showed that hGC-MSCs is normal diploid, no chromosome deletions or banding. Flow cytometry showed that hGC-MSCs were high positive for CD29, CD44and CD90, but low expressed or negative for CD45, CD34and HLA-DR. In vitro hGC-MSCs can successfully differentiate into lipocyte cell and osteoblast cell in differentiation medium. CONCLUSION There are hGC-MSCs in human gastric cancer, and their biological characters are similar as the normal human adult bone marrow-derived MSCs. hGC-MSCs may participate in the occurrence and development of gastric cancer, to isolated and cultured hGC-MSCs can be a valuable subject for gastric cancer research and for mesenchymal stem cell clinical application research.
