Objective:To study the cytotoxicity of cytokine-induced killer cells (CIK) combinedwith cisplatin on human lung adenocarcinoma cell line A549,to explore if there is asynergistic effect of them.Methods:In vitro, the healthy human peripheral blood mononuclear cells (PBMC)were induced into CIK cells possessing cytotoxicity with cytokines.CIK、cisplatin and CIKcombining cisplatin incubated A549cells for24h、48h, MTT assay was used to detectabsorbance values of each group cells and cell killing rate was calculated. Completeculture medium incubates CIK for48h, then diluted the supernatant with the same volumeof complete culture medium, incubated A549cells for24h with the diluting medium, A549cells apoptosis rate was detected by flow cytometry.Results:CIK incubated A549cells with target ratio5:1、10:1、20:1、40:1for24h、48h,A549cell killing rate was (19.11±1.89)%,(25.40±4.14)%,(39.41±2.73)%,(50.65±5.30)%and(21.09±5.21)%,(43.87±1.90)%,(50.21±4.06)%,(66.17±1.92)%respectively.Statistical analysis showed that the target ratio of5:1for48h, A549cellkilling rate has no significant difference than the same target ratio. When target ratio was10:1,20:1,40:1and co-cultured for48h, A549cell killing rate was significantly higher than24h (P <0.05);At the same time, CIK killing effect on A549increases with the ratio (P<0.05). Cisplatin with concentration of2.5,5,10,20,40μg/mL incubated A549for24h、48h, A549killing rate was (10.19±1.56)%vs(27.90±2.69)%,(33.42±5.37)%vs(49.85±2.48)%,(42.34±4.69)%vs(65.69±1.35)%,(49.85±3.84)%vs(72.18±1.06)%,(55.89±3.43)%vs(79.36±1.17)%respectively. The result showed that the killing rate of cisplatinon A549increased with the concentration at the same time(P <0.05).The killing rate ofcisplatin on A549with concentration of5,10,20,40μg/mL at48h was significantly hiherthan24h (P <0.05).5μg/mL of cisplatin combined with target ratio of10:1,20:1CIKco-cultured for24h, A549killing rate was (53.80±6.81)%,(61.17±3.82)%, higher than cisplatin or CIK alone (P <0.05). CIK supernatant can induce apoptosis ofA549cells.Conclusion:CIK enhances cisplatin cytotoxicity on lung adenocarcinoma cell lineA549in vitro.
