Objective:This experiment using e. coli k-12COL+to extract the e. coli E2、lactobacillus acidophilus extract Lactocidin, discusses two kinds of extracts to pathogenic e. coli and drug resistance of e. coli antibacterial effect in vitro; and the production of urinary tract infection rabbit model, respectively make pathogenic escherichia coli urinary tract infection rabbit model and drug resistance of e. coli infection of rabbit model, To observe two different bacteriocins the therapeutic effect of two kinds of bacteria model.MethodsThe first part:Using ammonium sulfate precipitation method to extract colicin E2and lactocidin, Yang’s method is used to extract colicin E2and lactocidin. Using SDS-PAGE gel to the molecular weight identified to determine.The second part:Use Kirby Bauer-disk diffusion method (K-B) bacteria medicine sensitive experiment, determine the best optimal inhibitory concentration of the extract.The third part:Production of rabbit urinary tract infection model,30rabbits were divided into five groups, group A control group, group B pathogenic E. coli using colicin E2treatment, group C pathogenic E. coli using Lactocicin treatment, group D resistance E.coli using colicin E2treatment, group E resistance E.coli using Lactocidin treatment. in1、3、5、7and10days to take urine culture medium plate counts observed effect of Lactocidin and colicin E2therapy drug resistance Escherichia coli and pathogenic Escherichia coli effect.result: The first part:Using ammonium sulfate precipitation method can successfully extract Lactocidin and colicin E2, and from the SDS-PAGE gel clearly see its beaches with; using Yang’s method can be successfully extracted colicin E2and Lactocidin and clear to see its beaches tape from the SDS-PAGE gel.The second part:Using the K-B method detected colicin E2the best inhibitory concentrations is64mg/ml,Lactocidin the best inhibitory concentration detected as8mg/ml.The third part:Using PH3diluted hydrochloric acid5ml via catheter injection of rabbit bladder5minutes can successfully establish a rabbit model of urinary tract infection. Colicin E2and lactocidin element on the pathogenic e. coli and e. coli resistance after bladder perfusion urine plate in SPSS statistics, the P<0.05, with statistical significance.Conclusion:The ammonium sulfate precipitation method and the Yang method can successfully extract colicin E2and lactocidin. By combining the analysis of E. coli in vitro antibacterial tests and animal experiments colicin E2on pathogenic E. coli and resistance E. coli have good antibacterial effect, lactocidin inhibitory effect on pathogenic E. coli better, but at resistance E. coli have no antibacterial effect.
