| ObjectiveUse the model of MCAO-reperfusion was induced by the method of monof ilament blocking the internal carotid artery, studied the role of prevention and treatment of Compound Xueshuantong Capsule on ischemic stroke. Provided ph-armacological evidence for the prevention and treatment of ischemic stroke for the cl-inic application.MethodsThe SD male rats were randomly divided into7groups:sham group, model group, Nimodipine group, Naoan capsule group, low dose group of Compound Xueshuantong Capsule (0.375g/kg),medial dose group of Compound Xueshuantong Capsule(0.75g/kg) and the large dose group of Compound Xueshuantong Capsule(1.5g/kg), intragastric administration,once a day, consecutive7days. After the last time of medicine, SD rats were anesthetized with10%chloral hydrate (i.p.), a small incision was made on the external carotid artery (ECA), the monofilament was inserted through the incision into the right internal carotid artery (ICA) and then into the origin of the MCA, in order to block the blood supply of ipsilateral middle cerebral artery in all.2h later, the monofilament could be drawn out when reperfusion was performed.The operation of sham group as same as operation group, except inserting the the monofilament. Postoperative10h repeated intragastric administration for1time.Berderson neurological defect score was used to analyse the neuroetholog--y of rats after reperfusion4,10,24h. The brain electrical activity was recorded by PowerLab bioelectricity amplifying system; The infarction volume percentage of brain was tested by TTC staining; The water content of brain tissue was calculated by weighing body weight, take the whole brain, weighing dry weight and wet weight of brain;HE staining was used to detect the pathology of brain; In order to further study the mechanism,The cerebral microcirculatory blood flow was recorded by the doppler microcirculation analyzer; The content of Glu〠MDAã€NOã€Lacã€Pyru and LDH in brain were measured by the kits; The content of TXA2ã€PGIã€ET-1and CGRP in serum through the method of Enzyme-linked immune.Results.The experimental results show that:compared with sham group,the score of neurological deficits after reperfusion4,10,24h, the infarction volume percent age of brain, the water content of brain of model control group rats all were si gnificantly increased; the structure of neurone were destroyed.what’s more, the brain electrical activity, the cerebral microcirculatory blood flow and the content of PGI2and CGRP in serum all were significantly lessen.Compared with model group, the score of neurological deficits of low dose group of Compound Xueshuantong Capsule,medial dose group of Compound Xueshuantong Capsule and the high dose group of Compound Xueshuantong Capsule decreased significantly at the reperfusion4,10,24h; the brain electrical activity of low, medium and high dose group increased significantly; the infarction volume percentage of brain of low, medium and high dose group decreased significantly; the water content of brain of medium and high dose group decreased significantly; the pathological score of low, medium and high dose group were lessen.Further mechanism research show that, compared with sham group, the cerebral microcirculatory blood flow and the content of PGI2and CGRP in serum all were significantly lessen.The content of Gluã€MDAã€NOã€Lac,LDH, TXA2,ET-1and TXA2/PGl2,LPR were significantly increasedCompared with the model group, the cerebral microcirculatory blood flow of low, medium and high dose group increased significantly;the content of TXA2of low dose group of Compound Xueshuantong Capsule lessen significantly; the content of PGI2of high dose group of Compound Xueshuantong Capsule increased significantly; TXA2/PGI2of low dose group of Compound Xueshuantong Capsule lessen significantly; The content of ET-1of low dose group of Compound Xueshuantong Capsule lessen significantly; the content of CGRP of low,medium dose group of Compound Xueshuantong Capsule increased significantly; the content of MDA and NO of low, medium and high dose group decreased significantly; the content of Glu of the high dose group of Compound Xueshuantong Capsule decreased significantly; the content of Lac and LDH in brain and LPR of the high dose group of Compound Xueshuantong Capsule decreased significantly.ConclusionProphylactic administration of Compound Xueshuantong capsule can be an effective intervention in rats with middle cerebral artery occlusion-reperfusion injury,and the intervention mechanism may be related to the improvement of microcirculation, protection from free-radical damage, the antagonists of excita-tory amino acid toxicity and the improvement of energy metabolism. This exper-imental research has provided pharmacological evidence for the prevention and treatment of ischemic stroke for the clinic application. |
