| Purpose:Aquilaria sinensis is an important part of Lingnan local herb resources. Because of its precious medicinal value and considerable commercial value,resources of the Aquilaria sinensis is on the verge of extinction. Furthermore,Nowadays,the development and utilization of the resources of the Aquilaria sinensis basically limited to the medicinal parts (the heartwood,including the resin), but as evergreen arbor,Leafy parts having the highest levels,were often discarded,causing serious waste of resources.Therefore, My study was oriented towards anti-inflammatory efficacy of the leaves of Aquilaria sinensis(Lour)Gilg. Basis on the pharmacologically active site and the active ingredient of the leaves of Aquilaria sinensis (Lour)Gilg; using the chemical analysis and efficacy screening techniques, to study the correlation of the pharmacologically active site and the anti-inflammatory efficacy, to identify the material basis of anti-inflammatory efficacy of the leaves of Aquilaria sinensis(Lour)Gilg, and On this basis,establishing HPLCFingerprint of the leaves of Aquilaria sinensis(Lour)Gilg,preliminary study its quality evaluation criteria. To provide a theoretical basis for further development and utilization of the of resources of Aquilaria sinensis.Method:To study the efficacy material basis of the anti-inflammatory and analgesic of the leaves of Aquilaria sinensis (Lour)Gilg, from a variety of pharmacodynamic evaluation model in vivo,initially identifying of the effective parts of anti-inflammatory in the leaves of Aquilaria sinensis (Lour)Gilg.To study the effective part of anti-inflammatory of the leaves of Aquilaria sinensis (Lour)Gilg on LPS stimulated RAW264.7cells, using ELISA assay to explore anti-inflammatory activity of different positions of the leaves of Aquilaria sinensis(Lour)Gilg,on macroporous resin, from the expression of TNF-a and IL-6.Establishing HPLC fingerprints of effective site of anti-inflammatory in the leaves of Aquilaria sinensis (Lour)Gilg, marking the main chemical components of anti-inflammatory effective part, providing a theoretical basis to explore the anti-inflammatory substances in the leaves of Aquilaria sinensis, as well as studying its quality.Marking major components of different positions of the leaves of Aquilaria sinensis (Lour)Gilg on macroporous resin as the target, isolating and identifying these compounds, initially confirming the material basis of anti-inflammatory efficacy of the leaves of Aquilaria sinensis (Lour)Gilg..Using ultrasonic extraction technique,aim of the main active ingredient content of in-flammatory from the leaves of Aquilaria sinensis, using ethanol concentration, extraction time, solid-liquid ratio, extraction temperature and other factors as influencing factors to study the best extraction technology of the leaves of Aquilaria sinensis(Lour)Gilg.And using the content of Mangiferin as an indicator to explore the bestharvest season of the leaves of Aquilaria sinensis (Lour)Gilg.Results:The results showed that the leaves of Aquilaria sinensis has significant efficacy of anti-inflammatory, and its n-butanol part, was the active ingredient of anti-inflammatory efficacy. Further study showed that three positions on macroporous resin:10%ethanol,30%ethanol and50%ethanol of the n-butanolparts in the concentration range of safety, had different degrees of anti-inflammatory activity in vitro,and the efficacy showed a concentration-dependent effect. By tracking and identifying the HPLC fingerprint of n-butanol Parts, six major compounds in the these positions were isolated:2-O-a-rhamnoside-4,6,4’-trihydroxy benzophenone,3-C-glucoside-2,4,6,4’-tetrahydroxy benzophenone, mangiferin,3.5-C-glucoside-2,4,6,4-four dihydroxybenzophenone,5,4-dimethyl-7-hydroxy flavonoids, apigenin-7,4’-dimethyl ether, in which mangiferin had the largest content and was most stable, thus, mangiferin was chosen as the indicators of inflammatory ingredients of the leaves of Aquilaria sinensis (Lour)Gilg.Studies on the ultrasonic extraction technology and the bestharvest season of the leaves of Aquilaria sinensis (Lour)Gilg.The results showed that the best extraction process of mangiferin was as follows:70%ethanol, ratio of liquid to material20:1, after soaked10min and40min ultrasonic extraction, and the best harvest season of the leaves of Aquilaria sinensis (Lour)Gilg is July.Conclusion:In this study, the anti-inflammatory efficacy and effective site of the leaves of Aquilaria sinensis (Lour)Gilg. were used as the starting point, the utilize of chemical analysis and efficacy of screening techniques was used to mutually verify the correlation of effective site and anti-inflammatory efficacy. Studying the material basis of anti-inflammatory effects of Aquilaria sinensis. Ultimately,six chemical composition of n-butanolparts of Aquilaria sinensis was determined as its effective part of anti-inflammatory:2-O-a-rhamnoside-4,6,4’-trihydroxy benzophenone,3-C-glucoside-2,4,6,4’-tetrahydroxy benzophenone, mangiferin,3.5-C-glucoside-2,4,6,4(-four dihydroxybenzophenone,5,4-dimethyl-7-hydroxy flavonoids, apigenin-7,4’-dimethyl ether. And based on these, primarily studies its quality evaluation criteria This study not only provides a theoretical basis for further exploitation of the resources of Aquilaria sinensis, but also provides an experimental basis for the development of series of related products of the leaves of Aquilaria sinensis (Lour)Gilg. |
