Compare Research Of The Protective Effect On Experimental Intracerebral Hemorrhage (ICH) In Rats Treated By The Electro-acupuncture (EA) At Different Time

[Objective]To study the intervention of behavior,brain histomorphology, nestin andbrain-derivedneurotrophic factor after experimental intracerebral hemorrhage(ICH)in rats treated by the electro-acupuncture(EA) at different time points.[Methods]192rats were divided into normal control group,sham operated group,ICHmodel group and EA group.EA group was divided into6h,24h,3d and7d subgroupsby the beginning of EA treatment. Normal control group,sham operated group andICH model group were also divided into four groups.every subgroups contains12rats.With the method of injecting autologous blood to produce the ICHmodel,Berderson score to evaluate behavioral change,observe the change of each ratbrain tissue appearance by light microscope after the intervention, the proteinexpression of nestin was detected by immunohisto-chemistry, and the expression ofbrain-derivedneurotrophic factor was detected by in situ hybridization.[Results]1.Berderson score to evaluate behavioral change once more before killing allthe rats,there’s no statistical significance between the subgroups(P>0.05);2.It was discovered that the model set’s brain tissue surrounding the hematomawas necrosis,dropsy,contain lots of hemosiderin particles,karyopyknosis of nervecells,vacuolization and phlegmasia cells infiltrated with the method of observationunder light microscope.Compare with the model set,the EA set ’s damage wasabatement,especially of the beginning of6h treatment.Normal control group and sham operated group were relatively showed no significant d ifferences.3. Compare with the model set,the protein expression of nestin was obvious inthe EA set,and had the statistical significance(P<0.01). Among the subgroups ofthe EA set,the nestin expression of6h subgroup was obvious than the24h,3d and7dsubgroups(P<0.05).There was no statistical significance between with thesubgroups of the model set.There were no expression of nestin in the normal controlgroups and sham operated groups.4. Compare with the model set,the expression of brain-derivedneurotrophicfactor was obvious in the EA set,and had the statistical significance (P<0.01).Among the subgroups of the EA set,the nestin expression of6h and24hsubgroups were obvious than the72h and7d subgroups(P<0.05).There was nostatistical significance between with the subgroups of the model set.There were noexpression of brain-derivedneurotrophic factor in the normal control groups andsham operated groups.[Conclusions]This research was based on the establishment of experimental intracerebralhemorrhage(ICH) rats model,to observe the protective effect on ICH of thedifferent intervention time with an acupuncture treatment on point of Bai-hui andRen-zhong.The results suggest that electro-acupuncture(EA) treatment may raise theexpression of nestin and brain-derivedneurotrophic factor to protect the braintissue.The EA may raise the expression of nestin to promote the proliferation anddifferentiation of neural stem cells. The EA treatment at different time points canlessen the dropsy of the brain tissue,lighten the inflammatory reaction,protect thenerve cells and promote the function recovery of the brain tissue. The effect of ICHby EA treatment in the early time was best.
