| ObjectiveThrough acupuncture at FeiShu(BLI3),DaZhui(DUl4),FengMen(BL12) in asthma modelrats.Observe the acupuncture to asthma model in rat lung tissue and serum T-bet,GATA-3transcription factors protein expression and the influence of HE staining lung tissue pathology,further explore the mechanism of acupuncture and moxibustion treatment of asthma, providescientific theory basis for acupuncture and moxibustion treatment of asthma.Method40healthy male SD rats were randomly divided into acupuncture treatment group, drugtreatment group and model control group, blank control group, each group consisted of ten.Acupuncture treatment group, drug treatment group and model control group set up ratsaerosolized asthma model use the intraperitoneal injection of egg ovalbumin (OVA) andaluminum hydroxide dry powder configured to10%of the composite OVA and the1%of theOVA solution. Blank control group use0.9%normal saline instead of egg ovalbumin,both in theprocess of intraperitoneal injection and aerosol inhalation.Acupuncture treatment group selectFeiShu(double),DaZhui and FengMen(double).The drug treatment group intraperitoneal injectionof0.05%sodium solution dexamethasone.After treatment, emply the method of immunehistochemical to determine the expression of T-bet and GATA-3expression in lung tissue andserum.Results1.General changes in rats:rat model group compared with the blank group, occurring sneeze, qiangke, shortness ofneither Hale nor hearty, unresponsive. After acupuncture and drug treatment of rats significantlyreduce the symptoms of sneezing, wheezing, and responsive.2. Compare each rat lung tissue morphology:Model control group rats lung airway epithelium, mucosal thickening, a large number of inflammatory cells infiltration, lung tissue wide alveolar septa, alveolar wall cracks expand evenfusion; Acupuncture treatment group, the structure of the drug treatment group bronchus basicintegrity, lung tissue inflammation infiltration cells less, basic normal alveolar structure; Ratbronchial lung tissue structure complete blank control group, no significant loss and folds, noinflammatory cell infiltration, alveoli and alveolar interval structure of normal form.3. Each group T-bet protein expression in rat lung tissue:Model control group compared with blank control group, model control group oftranscription factors in lung tissue of rats T-bet protein positive expression significantly reduced,statistically significant difference (P<0.01); The drug therapy group and acupuncture treatmentgroup compared with model control group, T-bet protein positive expression of transcriptionfactors, statistically significant difference (P<0.01), is still lower than the blank control group;Acupuncture treatment group compared with the drug treatment group, the difference wasstatistically significant (P <0.01).4. Each group GATA-3protein expression in rat lung tissue:The positive expression of GATA-3in the lung organization of model group is increase,butthe acupuncture group is decrease,the values of (P<0.01) were considered statisticallysignificant;the drug treatment group compaired with the model group is significant (P<0.01); theacupuncture group compaired with the medicine is significant too (P<0.01).5. Group T-bet comparison of serum in rats:Model control group compared with blank control group, T-bet in rat serum proteinexpression significantly reduced, statistically significant difference (P<0.01); Acupuncturetreatment group and drug treatment group compared with model control group, T-bet proteinexpression increased significantly, the difference statistically significant (P<0.01), but still lowerthan the blank control group; Acupuncture treatment group compared with the drug therapy group,the difference statistically significant (P <0.01).6. Group GATA-3comparison of serum in rats:Model control group compared with blank control group, GATA-3protein expression inserum of rats significantly higher, statistically significant difference (P<0.01); Acupuncturetreatment group and drug treatment group compared with model control group, GATA-3proteinexpression significantly reduced, statistically significant difference (P<0.01), but still higher thanthe blank control group; Acupuncture treatment group compared with the drug therapy group, thedifference statistically significant (P <0.01).Conclusion1.Shao’s Method of Wu Zhen can effectively improve the behavioral symptoms of asthma model rats.2.Shao’s Method of Wu Zhen can make the asthma model of rat lung tissue andinflammatory cells in the blood decreases, can repair the damage of bronchial mucosal epithelium,shows that acupuncture can inhibit airway inflammation, improve the effect of airwayhyperresponsiveness, this may be a shaw "five stitch" one of the mechanism of action ofprevention and treatment of asthma.3.Shao’s Method of Wu Zhen can make the asthma model T-bet protein content in the lungtissue of rats increased significantly, still can make asthma model GATA-3protein content in thelung tissue of rats significantly reduced.4.Shao’s Method of Wu Zhen can make the asthma model T-bet protein content in serum ofrats increased significantly, still can make asthma model rats obviously decrease GATA-3proteincontent in serum.This study suggests that shaw Shao’s Method of Wu Zhen may by raising T-bet content inserum and lung tissue, reduce the lung tissue and serum GATA-3protein expression, so as toreduce the inflammatory cell infiltration, inhibit airway inflammation, control of asthma, foracupuncture and moxibustion treatment of asthma provides scientific theory basis. |
