Methods:50clean pregnant rats were randomly divided into5group Aã€Bã€Cã€Dã€E. Except control group, group Bã€Cã€Dã€E were injectedintraperitoneally35mg/kg streptozotocin (STZ)on the third day of pregnancy, thenormal control group were injected with the equal dose of citrate buffer. From the5th day of pregnancy, Cã€E group were intragastriced with folic acid0.4mg/kgï¼›Dã€E group were intragastriced with vitamin E50mg/kgï¼›and Aã€B group withsame volume of distilled water for7days. After nature birth, myocardialpathological changes were observed for the neonatal rats under light microscope;Immunohistochemical and western-blot were used to detect apoptosis in the heartof fetus. Results:1.The blood glucose of pregnant rats in the5groups showed no difference before molding (p>0.05).There is significant difference between groupB and Aã€group C and Aã€group D and Aã€group E and A(p<0.01), but nodifference among group Bã€Cã€Dã€E(p>0.05).2.Under light microscope, themyocardial tissue in group Bã€Cã€Dã€E showed different degree damage, necrosisã€edema, the myocardial fibers showed disorder and inflammatory cellsinfiltration.3.In the fetal hearts, the mean OD value of caspase-3in5groups hadsignificant deviation (p<0.01).4.Among5groups, the grey value of caspase-3protein expression showed significant difference of fetal rats cardiacmyocytes(p<0.01). Conclusions: Gestational diabetes mellitus can cause fetalcardiac myocytes apoptosis, vitamin E and folic acid supplementation duringpregnancy have protective effect on the apoptosis of fetal cardiac myocytes, folicacid has an advantage over vitamin E to inhibit fetal cardiac myocytesapoptosis, supplementation of vitamin E and folic acid meanwhile is superior tosupply alone one of them. | | Keywords/Search Tags: | vitamin E, GDM, Apoptosis, Rat, folic acid, cardiac myocytes | PDF Full Text Request | Related items |
