Objective: Patients with severe acute pancreatitis (SAP) always appeargastrointestinal dysmotility, even gastrointestinal function failure. The subject ofthis study was to investigate the changes of nitric oxide synthase (NOS) andvasoactive intestinal peptide (VIP) neurons in gastric antrum myenteric gangliaof SAP rats with gastrointestinal dysmotility.Methods: Twenty female Sprague Dawley rats were divided randomlyinto two groups: SAP group and sham operated group. The models of SAP wasinduced by bile duct infusion of5%sodium taurodeoxycholate. The rats insham operated group had sham operation without bile duct infusion.24h afteroperation, pancreatic lesion was scored. The gastric emptying rate was measured.The percentage of NOS neurons and VIP neurons in the total neurons in gastricantrum myenteric ganglia were calculated. The data were dealt with SPSS20.0.Results:1. The rats in SAP group displayed ascites and calcification inabdominal cavity. Pancreas showed hemorrhage and necrosis. There were gastricretention and intestinal dilatation.2. Compared with the sham operated group,the gastric emptying rate in the SAP group was significantly lower [(70.99±1.82)%vs (31.49±3.96)%, P<0.05].3. Compared with the shamoperated group, the score of pancreatic lesion in the SAP group was remarkablyhigher [(1.60±0.52) vs (14.63±1.06), P<0.01].4. Compared with the shamoperated group,the percentage of NOS neurons in gastric antrum myentericganglia in the SAP group was significantly higher [(15.46±2.86)%vs(24.30±3.12)%, P<0.01].5. Compared with the sham operated group, thepercentage of VIP neurons in gastric antrum myenteric ganglia in the SAP groupwas remarkably higher [(10.72±1.92)%vs (22.45±4.10)%, P<0.01].6. Thecorrelation for gastric emptying rate and NOS neurons was negatively correlated(rs=-0.830, P<0.01).7. The correlation for gastric emptying rate and VIPneurons was negatively correlated (rs=-0.880, P<0.01).8. The correlation forthe percentage of NOS neurons and VIP neurons was positively correlated(rs=0.785, P<0.01), the difference was statistically significant.Conclusions: A high expression level of NOS neurons and VIP neuronsin gastric antrum myenteric ganglia of SAP rats may be one of the nervousmechanisms underlying SAP complicated by gastrointestinal dysmotility, andNOS neurons and VIP neurons were in correlation.
