Objective: The POSTN and CPB1proteins were not reported the differentialexpression proteins of gastric cancer in our proteomics research. The expression ofboth were detected in process of gastric mucosal cancerization, then the clinicalsignificance of both expression was analyzed.The results will provide valuableexperiment datas for gastric cancer patients on clinical early diagnosis, clinicalpathology grouping and evaluation the clinical prognosis.Methods: The8cases normal gastric mucosa, cancer side tissue and gastric cancerwere respectively collected and extracted the total protein of different tissues. Theexpression of POSTN and CPB1proteins was checked by Western blotting indifferent tissues. Then, the expression of POSTN and CPB1proteins was detected byimmunohistochemistry staining in gastric tissue arrays included of normal gastricmucosa, cancer side tissue and gastric cancer. Last,the clinical pathology significanceof both expression was analyzed in process of gastric mucosal cancerization.Results:1. The Western blotting results displayed that the expressions of POSTN andCPB1proteins were higher in gastric cancer tissue than that in normal gastric mucosaand cancer side tissue (P<0.01). Both expressions were higher in cancer side tissuethan that in normal gastric mucosa (P<0.01).2. The immunohistostaining resultsshowed that the expression positive rates of POSTN protein were respectively14.71%(5/34),46.15%(12/26) and77.78%(63/81) in normal gastric mucosa, cancerside tissue and gastric cancer tissue. The expression positive rates of POSTN proteinwere respectively60.00%(3/5),70.59%(12/17) and81.36%(48/59) in gastric welldifferentiated adenocarcinoma, gastric middle differentiated adenocarcinoma andgastric poorly differentiated adenocarcinoma. The expression positive rates of CPB1protein were respectively88.24%(30/34),69.23%(18/26) and17.28%(14/81) in normal gastric mucosa, cancer side tissue and gastric cancer tissue. The expressionpositive rates of CPB1protein were respectively40.00%(2/5),23.53%(4/17) and13.56%(8/59) in gastric well differentiated adenocarcinoma, middle differentiatedadenocarcinoma and poorly differentiated adenocarcinoma.3.The expressin positiverate of POSTN protein was higher in gastric cancer tissue than that in normal gastricmucosa and cancer side tissue (P<0.01). The expression of POSTN protein washigher in cancer side tissue than that in normal gastric mucosa (P<0.01). Theexpression of POSTN protein was related with tissue differentiation level and theexpression positive rate was higher in gastric poorly differentiated adenocarcinomathan that gastric well differentiated adenocarcinoma and middle differentiatedadenocarcinoma (P<0.01). The expressin positive rate of CPB1protein was lower ingastric cancer tissue than that in normal gastric mucosa and cancer side tissue(P<0.01). The expression of CPB1protein was lower in cancer side tissue than that innormal gastric mucosa (P<0.01). The expression of CPB1protein was related withtissue differentiation level and the expression positive rate was lower in gastric poorlydifferentiated adenocarcinoma than that gastric well differentiated adenocarcinomaand middle differentiated adenocarcinoma (P<0.01).Conclusion:1. The expression of POSTN protein was higher in gastric cancer tissue than that innormal gastric mucosa and cancer side tissue. And its expression was higher incancer side tissue than in normal gastric mucosa tissue.But the expresssion ofCPB1protein was polarity with POSTN protein.2. The expressions of POSTN and CPB1proteins were related with effluence anddifferentaiation level of tissue of gastric cancer.
