| Objective In the progress of orthodontic treatment, various kinds of factors can lead to gingivitis inflammation,many past studies have showed that, there is close relationship between Fusobacterium nucleatum and periodontal disease or teenagers periodontitis.But there is little research between Fusobacterium nucleatum (F.nucleatum) or its virulence factors FadA adhesion element and gingivitis inflammation in the process of orthodontic treatment.This experience had used16SrDNA PCR technique to test120orthodontics treatment patients’ Fusobacterium nucleatum (F.nucleatum) or its virulence factors FadA adhesion element which are in gingival crevicular fluid(GCF),to discuss the relationship between they and the gingivitis inflammation in the process of orthodontic treatment,and analyzes its effect in the development of orthodontic gingivitis happening.Methods To establish16SrDNA PCR which is a kind of technology to test Fusobacterium nucleatum (F.nucleatum) or its virulence factors FadA adhesion element,use F.nucleatumATCC25586to test its sensitivity and specificity;also use this technique to test120orthodontics treatment patients’Fusobacterium nucleatum (F.nucleatum) or its virulence factors FadA adhesion element which are in gingival crevicular fluid(GCF),to analysis the relationship between it and GI.Results The16SrDNA PCR has high sensitivity and specificity,120Clinical samples,65amplified F.nucleatum were obtained,amongst, the detection rate of67.50%. The detection rate respectively is:38were from the orthodontic treatment group (69.09%),14from the normal control group (46.67%), and29from the periodontitis group(82.85%). Orthodontics treatment group showed apparent higher relevance ratio than the normal control group,periodontitis group showed higher relevence ratio than the orthodontic treatment group and the normal control group,there was significant variance among them (X2=9.756, P<0.01), use virulence factors FadA primer to direct amplicate FadA gene fragments which are in120patients,67amplified, positive rate was55.83%;32cases for orthodontics treatment group,10cases for the normal control group,25cases for periodontitis group, orthodontics treatment group showed apparent higher relevance ratio than the normal control group,periodontitis group showed higher relevence ratio than the orthodontic treatment group and the normal control group,there was significant variance among them(X2=63.53,P<0.01); Obvious positive correlation (rs=1.0000, P<0.01) were observed between GI (gingival Index)and the relevance ratio of Fusobacterium nucleatum (F.nucleatum) or its virulence factors FadA adhesion element of the treatment group by using spearman rank correlation analysis.Conclusion1. Fixed appliance which in orthodontic treatment can cause gingivitis inflammation, which can also cause GI increases.2. F.n ucleatum has been closely related to the pathogenic bacteria, which can also play an important role in the gingivitis inflammation hapeened in the progress of orthodontic treatment. |
